| Objective:This experimental study focused on the effect of Feifukang on mouse Lewis lung cancer, including the inhibiting rate,the cell cycle and the apoptosis rate, and the influence of Feifukang to the gene expression of p53,fas,fas-L,bcl-2 and bax.so as to provide scientific basis for research on clinic research.Method:First, establish Lewis lung cancer model in mice, then observe the growth state of the mice and tumor Inhibiting rate. The cell cycle and the apoptosis rate were measured by using the flow cytometer. use the RT-PCR to observe the influence of Feifukang to the gene expression of p53,fas,fas-L,bcl-2 and bax.Results:The growth state of Feifukang plus TAX group was better than TAX group, the Feifukang group was the best. The inhibiting rate of the high dose was 36%,higher than the medium dose and the low dose. Combination with the TAX was 60%(P<0.05), and the cell cycle analysis showed that the tumor cells of the high dose,Feifukang plus TAX and TAX at G1 stage and the apoptosis rate were higher than the other groups(P<0.05). The gene expression of fas,fas-L had no difference in the model group and the treatment group(P>0.05). The gene expression of p53 in the treatment group were higher than the model group, and the difference was significant except the low dose group(P<0.05). The gene expression of bcl-2 in the treatment group were lower than the model group(P<0.05). The gene expression of bax in the treatment group were higher than the model group, and the difference was significant(P<0.05).Conclusion:Feifukang can improve the growth state of the mice and decrease the adverse effects of TAX. It has a marked anti-cancer effect, and can block the tumor cells at G1 stage to regulate the cell cycle and induce the apoptosis of lung cancer cells. The effect of combining method is better than single therapy. While it can not only increase the bax gene expression and reduce the bcl-2 gene expression but also increase the p53 gene expression to induce the apoptosis of the tumor cells, this may be a mechanism to the therapy of the lung cancer. |
