Polysaccharide Adjustment Mechanism Of The Preliminary Studies On The Intestinal Flora In Mice

Objective:We prove that the 0.50g/ml polysaccharides from Radix Codonopsis has a good effect of adjustment throug the treatment of intestinal micro dysbiosis of mice with different concentration of polysaccharides from Radix Codonopsis.We cultivate the Bifidobacteria and E.coli in vitro,compare with the change of intestinal flora and acetic acid in vivo,to research the adjustment effects of polysaccharides from Radix Codonopsis on the intestinal micro dysbiosis .Methods:1. In vivo experiment ,(1)We take out 30 mice randomly as the normal control group,the rest of 110 mice as model group.we use the method of intragastric administration to the model group 0.3g/ml Lincomycin Hydrochloride,and to the normal control group 0.3g/ml normal saline.Both groups are twice a day for 3 days.We put 10 mice of model group and 10 mice of normal control group to death at the 4th day and detect if the model has successfully set up.We divide the rest of model group mice into 5 groups:three polysaccharides from Radix Codonopsis groups of different density,positive group, spontaneous recovery group.(2)We use the method of intragastric administration to treat the each group as 0.50g/ml correspondence Chinese materia medica,the positive group as 0.5 hundred million viable organism every ml ,the spontaneous recovery group and normal control group as 0.3g/ml normal saline,twice a day for 7days.We put 10 mice of each group to death at the 11th day and detect the quantity of four stains of flora which are Bacillus bifidus, Bacillus acidi lactici, enterococcus, enteric bacilli.We also detect the content of acidum aceticum and blood endotoxin and observe the state of enteric bacilli translocation to the liver and pathological recovery of intestinal mucosa.Results : 1.In vivo experiment:(1)The mice were intertinal micro dysbiosis after intragastric administration with antibiotics.The content of Bacillus bifidus and Bacillus acidi lactici has obviosly decreased(P<0.01).The content of enterococcus and enteric bacilli has obviosly increased(P<0.01). The content of acidum aceticum which is the metabolic product of anaerobic bacterium has obviosly decreased(P<0.01). The content of peripheral blood endotoxin has obviosly increased(P<0.01).After the drug treatment for 7 days,The content of bacterium and acidum aceticum in intestinal tract has both increased in the spontaneous recovery group, but lower than 0.50g/ml polysaccharides from Radix Codonopsis group and normal control group(P<0.05). The content of bacterium translocation to the liver and the endotoxin were more than 0.50g/ml polysaccharides from Radix Codonopsis group and normal control group(P<0.05). 0.50g/ml polysaccharides from Radix Codonopsis group mice pathological recovery of intestinal mucosa were obviously better than the spontaneous recovery group.The polysaccharides from Radix Codonopsis can foster the growth of normal flora,increase the content of volatile fatty acid in the intestinal tract, decrease the content of blood endotoxin, effective control the bacterium translocation to the liver and promote recovery of intestinal mucosa.Each index has Significant difference compared with spontaneous recovery group.2. In vitro experiment:There was no effects of polysaccharides from Radix Codonopsis on the growth of E.coli in vitro(P>0.05), but positive effects on the growth of Bifidobacteria in vitro(P<0.05),acetic acid of the Bifidobacteria metabolism was positively correlated with the numbers(P<0.05).The Bifidobacteria can effect the growth of E.coli in vitro(P<0.05).Conclusion:(1). The 0.50g/ml polysaccharides from Radix Codonopsis have an good adjustment effect for the mice of intestinal micro dysbiosis which was induced by the antibiotic.Each index of this experiment has indicated that middle density of polysaccharides from Radix Codonopsis was a little better than the other two concentration groups. (2) There was no effects of polysaccharides from Radix Codonopsis on the growth of E.coli in vitro, but positive effects on the growth of Bifidobacteria in vitro,acetic acid of the Bifidobacteria metabolism was positively correlated with the numbers.The Bifidobacteria inhibit the growth of in vitro. (3) Polysaccharides from Radix Codonopsis which is a good microecological modulator can improve the antagonism of Bifidobacteria to the other pathogenic bacteria in colon through advancing the acetic acid of the Bifidobacteria metabolism, strengthening the colonization resistance.
