Objective:Radix Aucklandiae is the dry rhizome of Aucklandia lappa Decne. It has the efficacy of promoting the circulation of qi, relieving pain, invigorating the spleen, helping digesting. ( 2010 Edition ) emploies paper-roasted Radix Aucklandiae, but wheat bran roasted Radix Aucklandiae acts the development and substitution for the former in the practical application.The specific technology standard of processing cannot confirm yet. Thus this topic is aimed at optimizing the processing technology of Radix Aucklandiae,comparing the changing role of processing, drafting the quality standards, by scientific and rational experiment.Material and method:1.Material:Radix Aucklandiaes were purchased from the Menghai Research Institute of Agriculture in Xishuangbanna,Yunnan province. Zhai yanjun Professor of pharmaceutical college identified them as the genuine.2.Method:(1) Using HPLC to compare the different technologies of processing for the index components'influence,employ the orthogonal test L9(34)based on the wheat brans'adding amount, processing temperature, and processing hours as factors to optimize the best one, with the contents of volatile oil, costunolide and dehydrocostuslactone as indexes.(2) Using HPLC and GC-MS to test and analyse the chemical composition of essential oil.(3) Study the changes in the antidiarrheal role of drugs's processing, using ink-boosting in small intestine of mice and giving them folium sennae decoctum to grow"large intestine lax mice".Observe the change in the anti-ulcer effect of crude drugs'processing, after giving the mice reserpine and forming"the acute ulcer stomach mice"mold.(4) Draft the quality standard of crude drug and bran-roasted one, according to assaying their 10 batches saled samples'impurity, moisture content, ash content, acid insoluble ash content, extract etc. and components'content.Results:1.The content of total essential oil, costunolide and dehydrocostuslactone were all decreased after processing. The optimum processing technology was decoction pieces stir-frying with 30% wheat brans for 10 minutes at 110~120℃. 2.Using HPLC and GC-MS, we determine the content of index components of volatile oil. The result was that the content of dehydrocostuslactone and costunolide decreased after processing with wheat bran roasted. The decoction pieces show 76 peaks, and evaluate 34 components; while processed pieces show 63 peaks, evaluates 22 components with GC-MS.3.Processed pieces of Radix Aucklandiaes can restrain ink-boosting in small intestine, control the large intestine lax effect of mice induced by folium sennae decoctum. The efficacy of high-dose group of processed was the strongest, while the low-dose group of decoction pieces was the weakest. Meanwhile, Radix Aucklandiaes can antagonize acute gastric mucosal injury effect of mice by reserpine, and obviously reduce ulcer index. The efficacy of high-dose group of processed was the weakest, while the high-dose group of decoction pieces was the strongest.4.Initially drawing up the quality standard of crude and bran-roasted one, that is, according to dry product calculating, amounts of costunolide and dehydrocostuslactone in Radix Aucklandiaes shall not be less than 4.0%, and in bran-toasted one shall not be less than 3.5%.Conclusion:1.Optimum processing technology of Radix Aucklandiae by the orthogonal test was obtained logicaly and availably.2.After processing the crude Radix Aucklandiae, the chemical composition of essential oil appeared only quantitative change, and we did not discover the new composition yet.3.Crude Radix Aucklandiae and bran-roasted one all have antidiarrheal and anti-ulcer effects. The former's anti-ulcer is stronger, but antidiarrheal role is weaker contrarily.4.Quality standard of both of them are stable and feasible.
