Objective To explore the effects of swimming exercises with different time on muscles of rat, the changes of Des and Vim immunhistochemistry and ultrastructure were observed in CM, SM and SMM after rat's swimming exercises with different time.Method After exercises of SE, SCDE and EST, rat's CM, SM and SMM were cut off. These specimens were treated respectively with regular procedures of paraffin sections and transmission electron microscope(TEM). The paraffin sections were stained by Des and Vim immunhistochemistry. The ultrastructure of muscles were observed by TEM.Result (1)CM: Des and Vim of CM continuously decreased from OH to 24H after SE. And the injury changes were found in CM' ultrastructure. 48 hour after SE, Des and Vim of CM began to increase and the injury changes of ultrastructure improved. This reseached also found that Des and Vim of CM continuously decreased to the lowest point and the injury changes of CM' ultrastructure were worst in 3H after last swimming of SCDE. And 24H after last swimming of SCDE, the changes of Des, Vim and ultrastructure improved in CM. After EST, Des, Vim and ultrastructure in CM of LIT had no significant changes compared with ETC. But Des and Vim of CM increased obviously and positive changes were loud in ultrastructure after MIT. After HIT, Des and Vim of CM increased significantly compared with ETC. And the injury changes were occurred in ultrastructure. (2)SM: The results showed that Des of SM continuously decreased to lowest point from OH to 24H after SE. Vim of SM increased and the injury changes were found in ultrastructure. 48 hour after SE, Des began to increase and Vim increased to the highest point. Meanwhile the the injury changes of ultrastructure were alleviated.. After SCDE, the change of Des and Vim of SM was similar to SE. In 8-week EST, Des, Vim and ultrastructure in SM of LIT had no obvious changes compared with ETC. In MIT, Des and Vim of SM increased significantly and the ultrastructure of SM had positive changes. In HIT, Des decreased and Vim increased. Meanwhile the ultrastructure had injury changes. (3)SMM: The results showed that Des, Vim and ultrastructure of SE had no significant changes compared with EC. But after SCDE, Des and Vim of SMM decreased and ultrastructure had injury changes. In 8-week EST, Des, Vim and ultrastructure in SMM of LIT had no significant changes compared with ETC. In MIT, Des and Vim of SMM increased obviously and the ultrastructure of SMM had adaptable changes. In HIT, Des and Vim decreased remarkably. At the same time, the injury changes were found in ultrastructure of SMM.Conclusion (1)Des and Vim are the essential ingredients of the muscle cell, which is important to maintain cell's constructure and function; and both the decrease and increase of IF can change the ultrastructure of muscle cell; (2)Swimming exercises with different time will have different effects on IF and ultrastructure of the muscle; both the decrease and increase of IF, the ultrastructure of muscle cell have different change. (3) IF and ultrastructure of muscle cell have different change on the different time after swimming exercise.
