The reality of low level biologic potency and micronomicin constituent always exists in the industry of micronomicin fermentation using micromonospora purpura in fermentation broth.Program for this problem, amino acid precursors,metal ion ferment regulator and hydrolyzed fermentation medium by protease are researched in this article which can effect the process of micronomicin fermentation. The main work and research results are as follows:1 Respectively added certain concentration glycine, lysine, glutamic acid, tyrosine, valine, leucine, isoleucine, methionine in the fermentation medium to research the ability change of synthesis micronomicin by micromonospora purpura in shaking flask culture. The results showed that glutamate can significantly enhance the biological potency of the fermentation broth. By optimizing of glutamic ratio in the fermentation medium founded that adding 0.5% glutamic in the fermentation medium, the fermentation potency of Micronomicin increased 300U. In a 75t fermenter,0.5% ratio of glutamate can make the biological potency of fermentation broth to increase 15%.2 Using shaking flask fermentation to research the ability of synthesis micronomicin by micromonospora purpura under the conditions with different concentration of Zn2+, Mg2+and Mn2+in the fermentation medium. The results showed that the Mn2+has a positive effect on biological potency of the fermentation broth. Under the 0.6μg/mL of Mn2+concentration, the biological potency of fermentation medium could be increased 339U.3 The cooperation effect of Mn2+and Glu on the ability of synthesis micronomicin by micromonospora purpura has been tested in shaking flask fermentation. The results showed that the best group only increased biological potency 188U, and which is lower than that of Mn2+,and Glu respectively effect reached. 4 In the selection and concentration optimizing of HT-type and HY-type fermentation regulator, Flask experiment results showed that the biological potency has increased 400U with 0.04% ratio of HY-type regulator in the fermentation medium.
