Along with society's development and progress, food safety attracts more and more attention. Because of the 2008's milk powder events, melamine becomes the focus of public security. Melamine and its homolog cyanuric do great harm on people's health and life safety, so, for these illegal additives, we need develop corresponding technical standard about fast detection and conclusive evidence in order to provide effective technical support for the people's health and food safety issues.Melamine, short for three amine and honey amine etc, it is a kind of triazine heterocyclic organic compounds. It is an important organic chemical raw materials, widely used in plastics, coating, adhesive, food packaging materials, disinfectants, fertilizers and pesticides production. Melamine in usually relatively stable, but at high temperatures it could decompose and emit cyanide. In acid or alkali environment, melamine happens hydrolysis, its amino gradually been replaced by hydroxyl, and generats its homolog ammeline, ammelide and cyanuric acid (The scientific name is 2,4,6-thihy-droxy-l,3,5-triazine). Melamine and cyanuric acid can combine closely to form insoluble melamine cyanurate, and precipitated in renal cell, then formed kidney stones. The available evidence suggests that ingestion of melamine separately may produce low toxicity, but ingestion of melamine and cyanuric acid together can lead to kidney toxicity, which affects human health seriously. The United States, the European Union and China has all banned melamine and cyanuric acid added to foods. Currently, the existing detection methods for simultaneous determination of melamine and cyanuric are U.S. FDA method and domestic GC-MS method and HPLC-MS method, but GC-MS method still need silicon alkylation derivative, the steps relatively complicated, the analyzed sample matrix include milk and dairy products and pet food. There are not reports about adopting LC-MS method to simultaneously detect melamine and cyanuric in milk and dairy products, including dairy products, animal-derived food and plant-derived food at this time. So, this paper developed a HPLC-MS/MS method and sample pre-treatment method for the determination of melamine and cyanuric residue in food. The sample was extracted with acetonitrile: water (1:1), and cleaned up with MCT SPE cartridge. The analyst was determined by HPLC-MS/MS at multi-reaction monitoring mode(MRM), was employed for the quantitative determination, and quantified by the internal standard curve. The separation was performed on an Atlantis HILIC Silica column with acetonitrile: lOmmol/L ammonium formate (9:1) and acetonitrile (containing‰1 formic acid)as mobile phase. This method make full use of the liquid chromatographic separation ability and mass spectrometry qualitative ability, it is simple, quick, accurate and suitable, the sample purification effect is good, the limit of detection can satisfy the limited requirements.
