Mulberry (Mulberry),scientific name Morus alba L, fasten moraceae, mulberry genus nearly mature JuHuaGuo mulberry compaction, say again mulberry, mulberry jujube, its fruit oval black or dark purple. Flowering 4~5 months; fruiting in June or July, mulberry usually harvested in 4~6 months,which is red and purple.The mulberry resources are very rich in China, due to the fresh mulberry easily decay, and relatively vulnerable hidden and storage,therefore squeezing juice is the most important processing method. The mulberry fruit juice after slag, seeds and scraps of still contains abundant nutritional and functional components, so extracted antioxidant substances from mulberry dregs was a kind of comprehensive utilization of mulberry dregs resources effectively.In the study, polyphenols were extracted from the mulberry dregs ,which from waste reuse in comprehensive use of resources, and takes a hydration gallic acid as standard reference substance. Using the Folin-Ciocaheu colorimetric determination of mulberry dregs of total polyphenols, its total polyphenol concentration and absorb light value of the linear regression equation for A y = 0.1705 x + 0.0336 (including y is absorbency, x for gallic acid density of apmsĪ¼g/mL), correlation coefficient of linear 0.9953, which show A good linear relations.Different extraction methods were analyzed,which got polyphenol compounds from mulberry dregs, and the order of the total polyphenol content was Ultrasonic assisted extraction > Microwave assisted extraction > Soxhiet extraction >Supercritical CO2 extraction. The total polyphenol content with ultrasonic assisted extraction was higher than other 3 methods, and the time required for short, suitable for the extraction of natural substances. Raw material pretreatment in different ways, the total polyphenol content was also different, mulberry dregs by vacuum freeze-drying got total polyphenol content was obviously higher than vacuum drying processing of raw material. The optimization extraction process of total polyphenols with ultrasonic assisted extraction in mulberry dregs was studied. Some factors influenced polyphenols extraction such as the ethanol concentration, the material-liquid ratio,the amplitude, the ultrasonic time. The single factor and orthogonal tests were conducted.When extracted the total polyphenols with ultrasonic assisted extraction from mulberry dregs,the order of each factor to the total polyphenols extract effects as follows: the amplitude>material-liquid ratio>ultrasonic time>ethanol concentration. And the results showed that the optimal extraction conditions as follows: materials mesh size was 80, pulse (40s, 40s), the material-liquid ratio was 1:70,the ultrasonic time was 160s,the ethanol concentration was 60%,amplitude was 90%, extraction two times. The extraction rate of mulberry dregs total polyphenols could get 73.3284mg/g.D101, AB - 8, NKA - 9, D4020 four kinds of macroporous adsorption resin in static adsorption and desorption test of special was evaluated, the result showed that the type NKA-9 macroporous adsorptive resin for a better separation of polyphenols compounds from mulberry dregs. The optimum absorption conditions of macroporous resin NKA-9 were as follows: concentration of feeding 1.61mg/mL, pH 4.5, feeding rate 2BV/h. The optimum desorption conditions are: elute-ethanol concentration 70%, elution rate 2BV/h, elution volume 3BV. Under above stated conditions, the obtained polyphenols purification was concentrated to 69.62%, while the polyphenols content was not purification 31.72% .So mulberry dregs polyphenol compounds coarse extraction liquid through macroporous resin purification, large quantities of impurities were removed, the content of polyphenol compounds were improved.The antioxidant properties of polyphenol extracts were used to reviewed in mulberry dregs, and compared with Vc and BHT. In this paper,the results showed that the polyphenol extracts from mulberry dregs had antioxidantt function. In a range of specified concentration, the total reductive ability of polyphenol compounds from mulberry dregs,which increased with increasing concentration. The results show that at a certain concentration,the order of T-AOC from high to low was purification liquid>crude liquid>Vc,and the inhibition ability on hydroxyl radical ranked from high to low: purification liquid>crude liquid>BHT.So the crude polyphenol compounds and purification polyphenol compounds from mulberry dregs both had strong antioxidative effect. The determination of polyphenol purification from mulberry dregs with the bacteriostatic circle diameter and minimum bacteriostatic concentration (MIC) ,which were studied the bacteriostatic effect.The results showed that mulberry dregs purified polyphenol concentration was 0.3% with E.coli ,which had the most obvious inhibitory effect, the E. coli inhibition zone diameter was14.43mm. Similarly, When mulberry dregs purified polyphenol concentration was 0.3%, the inhibition of Staphylococcus aureus was most obvious at this concentration, the zone of inhibition of Staphylococcus aureus diameter was 13.13mm.Mulberry dregs purified polyphenols inhibited Escherichia coli and Staphylococcus aureus,the minimum inhibitory concentration was 0.00625%. Our study can provide a frame of reference with research and industrial production for polyphenol extraction, purification and functional activity from mulberry dregs.
