L-Carnitine,which is a new kind of functional food additives,has many Physiological functions,such as accelerating energy metabolism, antifatigue. And it is widely used in medicament,foods and functional foods. On the contrary,D-Carnitine,its enantiomer,shows toxic and side-effect.Therefore,it's necessary to separate and determine D/L-Carnitine.The condition of pre-column derivatization of D/L-C and the HPLC method for determination are studied.Using (+)-FLEC as derivatization reagent, chromatographic condition:Agilent SB-C18 column,4.6mm×250mm,5μm; mobile phase: triethylamine phosphate buffer and acetonitrile (73:27, V/V); flow rate:1ml/min;260 excitation,310 nm emission monitoring; column temperature:25℃. Good separation effect, high-sensitivity and good stability of system and derivatives are demonstrated in this method. On that basis, the method for pretreatment of D/L-C determination in milk powder is studied,and pre-column derivatization -HPLC can qualitative and quantitative analysis.Using L-ALA-β-NA as derivatization reagent, mobile phase of acetonitrile: tetrahydrofuran:sodium dihydrogen phosphate (50Mmol/L, PH=2.6)=9:6:85 (volume ratio), and detector for the UV detector at wavelength of 244nm, flow rate: 1ml/min; column temperature:30℃. On that basis, the method for pretreatment of D/L-C determination in functional foods is studied,and pre-column derivatization-HPLC can qualitative and quantitative analysis.
