Mycotoxin is the secondary metabolic product of toxicogenic fungus, often widely existing in moldy grains. Once entering human's body, it may cause serious harm to human's health, like deformity, mutation or cancer and so on. Therefore, it has caused wide public concern in global food security over recent years. Consequently, it is of great significance to develop simple and effective analysis methods of mycotoxin to provide an effective guarantee for human health and food safety.A method was developed for the simultaneous determination of Aflatoxin (AFB,, B2, G1, G2), Zearalenone and Ochratoxin A in cereal grain by Liquid chromatography with fluorescence detection after immunoaffinity column clean-up and post-cloumn derivatization. Ground cereal samples were extracted with methanol-water (80:20,v/v), the extracts were purified by imunoaffinity columns and the toxins were separated by reversed-phase HPLC, and quantified with fluorescence detection after photochemical derivatization. The separation was performed on a WATERS Symmetry(?) C18柱(4.6 mm i.d.×150 mm,5μm)with a linear gradient of methanol-acetonitrile-1% phosphoric acid mixture. Recoveries of the different cereals (wheat, corn, rye) spiked with Aflatoxin (AFB1, B2, G1, G2), Zearalenone and Ochratoxin A at levels ranging from 0.24-6,4~100 and 0.5~40μg/kg were from 70.8% to 94.0% and relative standard deviations were between 2.79 and 9.38%, the limit of detection was 0.24,0.5 and 4μg/kg. The verification report results from 5 different laboratories showed that the established method is simple, rapid, accurate and applicable for simultaneous determination of Aflatoxin (AFB1, B2, G1, G2), Zearalenone and Ochratoxin A in cereal grain.
