Immunoaffinity column (IAC) is a simple, rapid, accurate separation method. A liquidchromatography-tandem mass spectrometry (LC-MS/MS) method and liquid chromatography(LC) method were established for simultaneous determination of six aflatoxins and sixzearalenone in milk and milk powder. The main results were as follows:(1) By optimizating the pretreatment methods, affinity column purification conditions,chromatographic detection conditions, IAC-LC-FD method of detecting AFs in milk and milkpowder is established successfully. This way the recovery rates for AFs in milk were between91.2%~113.8%, and the relative standard deviations were all less than9.80%, the limits ofdetermination were between0.002~0.04μg/L. The recovery rates for AFs in milk powderwere between88.4%~104.5%, and the relative standard deviations were all less than9.73%,the limits of determination were between0.008~0.016μg/kg.(2) By optimizating the pretreatment methods, affinity column purification conditions,chromatographic detection conditions, IAC-LC-UV method of detecting ZER and its agalogsin milk and milk powder is established successfully. This way the the recovery rates for ZERand its agalogs in milk were between54.22%~90.76%, and the relative standard deviationswere all less than9.44%, the limits of determination was0.05μg/L. The recovery rates forZER and its agalogs in milk powder were between61.0%~89.6%, and the relative standarddeviations were all less than8.42%, the limits of determination was0.15μg/kg.(3)By optimizating the pretreatment methods, affinity column purification conditions,chromatographic detection conditions, IAC-LC-MS/MS method of detecting AFs and ZERand its agalogs in milk and milk powder is established successfully. This way the the recoveryrates for AFs in milk were between92.7%~105.6%, and the relative standard deviations wereall less than7.12%, the limits of determination were between0.0016~0.003μg/L. Therecovery rates for ZER and its agalogs were between60.3%~86.3%, and the relative standarddeviations were all less than6.91%, the limits of determination were between0.008μg/L.This way the the recovery rates for AFs in milk powder were between91.9%~107.8%, andthe relative standard deviations were all less than9.86%, the limits of determination werebetween0.0075~0.01μg/kg. The recovery rates for ZER and its agalogs were between 62.4%~86.3%, and the relative standard deviations were all less than8.76%, the limits ofdetermination were between0.01μg/kg.
