Alternaria is one of fungus, existence in the feeds and foods. They can pollute crops,vegetables and fruits. It can grow in low temperature, so it can make fruits and vegetables moldy. Alternaria can produce many metabolites that consist of altenuene, alternario,alternariol monomethy,tentoxin and AAL etc. Some of metabolites can do harm to animals and plants, and some of them have cytotoxicity,teratogenicity and embryotoxicity. If people eat the foods that polluted with alternata, it will do harm to human health. And there have few reports about simultaneous detection many alternatas in fruits. In this paper, the extraction methods,purification methods and detection methods of Alternaria were researched. A high performance liquid chromatography with fluorescence detection method and an ultra high performance liquid chromatography tandem mass spectrometry detection method for simultaneous determination multiple altemarias in fruits were established. Furthermore,the new method was applied to real samples. The results show that the method meets the application in sensitivity and accuracy.This paper mainly include as follows:In Chapter1, the background and significance of thesis were introduced briefly. Including food safety problems,the hazards of mycotoxin and alternarias to people and advances in studies on alternarias.In Chapter2, established a method that could assay simultaneity altenuene^alternariol and alternariol monomethyl in fruits by a high performance liquid chromatography with fluorescence. Aalyte of the three altemarias in fruits were extracted with acetonitrile, then cleaned up by HLB and NH2solid phase extraction column, Analisis was performanced on a Chromolith Performance RP-18e column and determinated with a fluorescence detector. According to the above the test conditions, the result shows that, the average recoveries were between78.2%to103.6%with RSD between5.9%to8.6%. The method is effective simultaneous determination of altenuene,altemariol and alternariol monomethyl in fruits, The advantages of this method were easy, stability, good repeatability, prompt operation and stable and apprppriate for the demand of simultaneous determination of these three altemarias in fruits. In Chapter3, established a method that is simultaneous determination ALT, AOH,AME,tentoxin and AAL in fruits by ultra high performance liquid chromatography tandem mass spectrometry (UPLC-MS/MS). Fruits were extracted by acetonitrile and then was purificated with Oasis HLB solid phase extraction column, The analytical column was an Acquity UPLC BEH C18column, acetonitrile and water as mobile phase using gradient elution, and then analyzed using UPLC-MS/MS under the optimization of the mass spectrum. According to the above the test conditions, the result shows that, the average recoveries were between76.9%-113.4%with RSD between2.6%to5.5%. The advantages of this method were accurate qualitative and quantitative evaluation, better reproducible and high sensitivity. and apprppriate for the demand of simultaneous determination of these six alternarias in many fruits. Detection the content of alternarias with UPLC-MS/MS in mildewy fruits, and contrast the content of alternarias in mildew part and no mildew part in mildew fruit. Well-established that there also have the content of alternarias in no mildew part. It is great significance to prove that eat no mildew part in the mildewy fruit will also effect our health.
