Compound Pudilan is composed of four herbs of scutellaria baicalensis, radix isatidis, dandelion and viola yedoensis Makino,with good heat-clearing and Detoxicating, anti-inflammatory effect of swelling. It can be used to treat Furuncle, pharyngitis,quinsy, lymphadenitis, mumps etc,widely used in medicine, urology, surgery,eye,ear,nose,and throat. As the compound can occur through the function of ingredientsing together, restriction or introduction to achieve the goal of reducing poison and increasing effect. So it has its unique advantages for the treatment of inflammation.However,the current Pudilan preparation in saling mainly include anti-inflannat-ory table, oral liquid, anti-inflammatory capsule three basic forms,and they still do not get out of the stituation "black,big and bold"of the traditional Chinese medicine preparation. Just take the Pudilan anti-inflammatory tablets producted by Wuhan Dragon Pharmaceutical Co,Ltd for example,it needs4times a day,5-8tablets every time,needing too much dose,takeing frequently. It will cause great inconvenience to patients,and reduce patient compliance. Therefore,to overcome the above shortcomi-ngs of the preparation,get the popurse of "three small","three-way","five convenient",choosing to improve its technology, optimizing its extraction process, enhancing its active ingredients, then making it sustained-release preparation is certainly a good choice.This test is mainly key to the situation about extraction technology of production and clinical usage of Pudilan tablet of Wuhan Dragon Pharmaceutical Co, Ltd, trying to improve its extraction technology. Through the method of extracting it with water and depositing with acid,then solubling the deposition with alkali and depositing with alcohol,then depositing the leachate with acid again,we can extract and purify scutellaria baicalensis. Use the method of aluminum salt complex-in than color and HPLC to determinate the content of total flavone and baicalin in the extraction of scutellaria baicalensis respectively. Taking this for index,through single factor experiment combinating with orthogonal test,we findly get the best extraction and purification method:10times volume of water,extracting2times,1.5hours each time.The content of baicalin can up to88.01%,the content of total flavone can up to 93.55%.Extracting the radix isatidis with a certain concentration of alcohol,taking the content of findirubin determined by HPLC and alcohol-soluble extraction for indexs,through orthogonal test we determine the optimum extraction method of radix isatidis:9times the amount of50%ethanol-water solution,extracting2times,1.5hours each time. The content of indirubin and alcohol-soluble extraction can up to127.0989(μg/50g)and93.986%respectively. The method of determination is stable and possible.Extracting the dandelion with water,taking the content of caffeic acid determined by HPLC and rate of dry extraction for indexs,through orthogonal test we determine the optimum extraction method:10times the amount of water solution, extracting3times,1.5hours each time. The content of caffeic acid and the rate of dry extraction can reach to108.0021(μg/g crude drug),40.23%. The method of determination for caffeic acid is stable and possible.Verificating the above three optimization extraction method,the results indicate that the selected optimization methods have good reproducibility.
