Scutellaria baicalensis Georgi?SBG? is a perennial herb of the genus Astragalus,and it is a commonly used Chinese herbal medicine,which has the effects of clearing heat and dampness,purging fire and detoxification.It is also commonly used in the treatment of damp fever and upper respiratory tract infection.Due to its wide distribution,different planting years?xylem-rottened Scutellaria,xylem-norottened Scutellaria? and irregular processed?wine cellar,fried charcoal?,the quality of medicinal herbs and pieces is often uneven.In order to ensure the safety and accuracy of SBG,the quality control of medicinal materials is particularly important.At present,liquid chromatography?HPLC? fingerprinting method is used to compare the chemical composition and quantitative components to evaluate the quality of the drug.However,HPLC consumes long time and large solvent.For large sample analysis,the flux is limited.Ambient mass spectrometry?AMS?,as a new mass spectrometry technology have advantages of simple operation and fast analysis,which has been widely used in the quality control of Chinese herbal medicine.Paper-spray mass spectrometry?PS-MS? is one of AMS.It has real-time monitoring,simple and fast performance,and it is expected to develop a fast,reliable and real-time analysis method for quality control of traditional Chinese medicine.At present,there are few reports on the quality control of Chinese herbal medicines by PS-MS,and no reports of the analysis of SBG.In this thesis,the research work was carried out with xylem-rottened Scutellaria and xylem-norottened Scutellaria,and the specific content is as follows:1.Introduction.The main chemical components and pharmacological effects of SBG and methods and principles of establishing traditional Chinese medicine fingerprints were reviewed.The principle and application of paper spray mass spectrometry were introduced.The methods of processed and the influence of processed were summarized.2.Analytical study of xylem-rottened Scutellaria and xylem-norottened Scutellaria by HPLC.The fingerprints of them were established by HPLC.The optimal fingerprints were obtained by optimizing the extraction solvent and extraction method.The fingerprints of 9 batches xylem-norottened Scutellaria and 8 batches xylem-rottened Scutellaria were analyzed by ChemPatternTM chemometrics software.The results showed that the fingerprints of them had their own characteristics,and the difference between them was obvious.At the same time,high-performance liquid chromatography-mass spectrometry?HPLC-MS? was used to confirm the common components of them,and eight flavonoids were identified in the literature.Gas chromatography-mass spectrometry?GC-MS? was used to analyze the volatile components,and13 chemical components were determined by computer library and CAS number,which provided theoretical support for the development of medicinal resources of SBG.3.Ambient mass spectrometry was used to quickly identify xylem-rottened Scutellaria and xylem-norottened Scutellaria.The influencing factors of ambient mass spectrometry were optimized,and the PS-MS diagram of them was obtained.The paper base was modified with different metal salt solutions,and the difference in mass spectrum was increased by the complexation effect of metal and flavonoids.The chemometrics software analyzed the mass spectra of them,and found that the paper base modified by CuCl2,FeCl3 and CoCl2 can successfully identify them,and the stability of the CuCl2 modified paper base is better than the rest,which is the best experimental condition.The results of the PS-MS are consistent with the results of the HPLC,and it was proved that the method established by PS-MS can be used for the identification of xylem-rottened Scutellaria and xylem-norottened Scutellaria.4.Quality control of SBG processed based on HPLC and AMS.The baking method of SBG was explored.The medicinal materials was suffocated with wine for 24 hours,then dried in an oven at 95?,which is the best processed temperature.The mass spectrograms of the unprocessed products and processed products obtained by PS-MS method were analyzed by ChemPatternTM chemometrics software.The experiment established a rapid and accurate identification method of the unprocessed products and processed products,the 7 batches of the unprocessed products and processed products were clearly divided into two categories.
