E. coli and Salmonella are two of the world the most important food pathogenic bacteria. At present, the main treatment with antibiotics to E.coli and Salmonella infection, however many bacteria to antibiotics have become resistant. Therefore, the screening of low toxicity, no drug resistance of natural medicine as a new type of antibacterial agent become a research hotspot. It is reported that Chinese medicine Rhubarb and Scutellaria baicalensis have inhibitory effect on many kinds of bacteria. But for their inhibitory effect on E. coli and Salmonella of the main compounds and their antibacterial mechanism is unclear. Therefore,this study selected the extract from Rhubarb and Scutellaria baicalensis,designed to track these two kinds of traditional Chinese medicine(TCM)of E. coli and Salmonella antimicrobial active substances, and to explore its antibacterial mechanism and provide theoretical basis for the development of new type of antibacterial agent. This study on E. coli and Salmonella as the research object, using an hydrous ethanol extraction of Rhubarb and Scutellaria baicalensis, extract rotation generated byevaporation into concrete, in turn with petroleum ether, chloroform, ethyl acetate and positive.And then use the organic solvent extract of E. coli and Salmonella this two kinds of antimicrobial bacteriostatic experiment, selected the best extract bacteriostatic effect. By the experiments on rhubarb chloroform extract and ethyl acetate extract Scutellaria baicalensis part of E. coli and Salmonella has good inhibition, respectively by HPLC/MS/MS analysis rhubarb chloroform, ethyl acetate of Scutellaria baicalensis chemical composition. Determine the rhubarb chloroform extraction period of main compounds of aloe-emodin, rhein, emodin, chrysophanol and physcion.Ethyl acetate extract of Scutellaria baicalensis main compounds are baicalin, baicalein, wogonin and Wogonoside. Again respectively with these two extracts the main compounds of E. coli and Salmonella in the bacteriostatic test, the bacteriostatic effect of the best screening monomer.Rhubarb in chloroform extracts of E. coli and Salmonella bacteriostatic effect is the best monomer for rhein, effect of scutellaria baicalensis georgi best monomer for radix scutellariae.
