| As the head of the five Sourth China medicine (areca,villosum, fructu amomi, alpinia oxyphylla, balberd, tambac), areca is the second lagrest tropical crops which only rank after the rubber in Hannan Province. So far, the processing and application of the areca is maining on edible and officical. Except to eat in fresh, the traditional areca is mainly used in areca dry,some snack with spice, condiment etc, but the seed is always be discarded.This reserch regarding the seed of the Hainan areca as the object of research, separately adopting Ultrasound-microwave Collaborative Extration and Ultrasonic assisted Extration to extract the areca seed oil, meanwhile it come out a way of testing the extraction efficiency of the areca seed oil by Ultraviolet Spectrophotometry. In additional, this reserch analysis the oil's Ultraviolet spectrophotometry way and fatty acid composition, besides, it also processed a preliminary experiment of the acute toxicity. Ultraviolet Spectrophotometric Determination of areca seed oil extraction rateWith reference to the international standard GB/T14772-2008-betel nut oil,at wavelengths between200-300nm scan,standard oil solution of well prepared is measured at the location of its maximum absorption peak at210nm. Ultrasonic-assisted extraction of extraction of betel nut oilUsing the technology of ultrasonic of extraction of areca seed oil, One-factor experiment and Response surface experiments examine the extraction time, feed ratio, ultrasonic frequency on the effect of areca seed oil extraction rate to determine the optimum extraction conditions for:liquid ratio10:1, extract24min,28KHz ultrasonic frequencies, extraction rate of12.56%. Compared with the traditional method, extracting time greatly reduced. It is a better process for extraction of areca seed oil.3. Ultrasonic/microwave assisted extraction of betel seed oilThe use of Ultrasonic-microwave-assisted Extraction of betel nut oil,according to the Box-Behnken test and on the basis of single-factor test and betel nut seeds oil extraction rate of response, it adopts the three factors of by response surface analysis.Results indicate that the thermostat mode ultrasonic rated power50W,22min, temperature of65℃, liquid extraction time than11mL/g, areca seed oil extraction rate of up to14.29%. GC-MS analysis and ultrasonic assisted extraction of areca seed oil contains13types of fatty acids, oils containing unsaturated fatty acid content in45.84%; ultrasonic-microwave-assisted extraction of areca seed oil contains11kinds of fatty acids, which contains unsaturated fatty acid54.49%, two were detected linoleic acid at the first time.5. Toxicological StudiesOn the basis of preliminary experiment,"maximum dosage method" of SD rats by intragastric administration of acute toxicity tests, after delivery, animals showed no obvious toxicity symptoms, weight and blank control group no significant impact. Major life organ profile detection is no exception. No significant symptoms of acute toxicity and target organ toxicity.Paper for the first time with ultraviolet Spectrophotometric method for determination of content of betel nut oil, with response surface methodology to optimize ultrasonic extraction and ultrasonic-microwave synergistic extraction technology of areca seed oil, areca seed oil extracted in two ways to the physic-chemical properties and fatty acid composition of comparative analysis, and make the initial acute toxicity tests.
