Microwave-assisted extraction and ultrasonic-assisted extraction methods arenew fast, cheap, efficient method to extract dioscin. However, microwave extractionhas a low concentration gradient which restricts the extraction yield of dioscin, andultrasonic extraction method doesn’t product strong heat effect in cell as themicrowave method. An aqueous two phase extraction technology is the modernefficient separation method, having an advantage of keeping biological activity.Dioscorea zingiberensis. C. H. Wright contains many high medicinal value materialssuch as dioscin, deltonin, protodioscin, trillin, gracillin, which are worth extracting. Anovel method of ultrasonic-microwave auxiliary aqueous two-phase synergistic toextract total saponins of dioscorea zingiberensis. C. H. Wright has been proposed inthis research taking the advantages of methods mentioned above. Specific researchwork contents are as follows:(1) Dominant factors were determined by the investigation of the influence ofparticle size, soaking time of dioscorea zingiberensis. C.H.Wright, ultrasonic powerand other factors on the extract yield of total saponins with continuous variablemethod to study utrasonic-assisted solvent method. Based on the comparison of thetotal extraction yield of the traditional method, ultrasonic auxiliary solvent methodand microwave auxiliary solvent method, the experiments shown that the microwaveaided method and the ultrasonic auxiliary method have a higher extract rate. And theexperiments also reveal that the there were higher extraction efficiency whenmicrowave auxiliary solvent extracting with stirring and ultrasonic auxiliary solventextracting in the static situation.(2) The extraction of total saponins in dioscorea zingiberensis. C. H. Wright wasinvestigated by two-aqueous of phase system of ethanol-dipotassium hydrogenphosphate auxiliary with microwave irradiation on the utilization of orthogonalexperiment. Based on the investigation of the main influence factors on the totalsaponins extraction, optimization conditions were determined. The optimizationconditions were the particle size of80meshes, microwave power of390W andmicrowave irradiation for5min. The factors that will influence the extraction werelisted: the particle size> microwave time> microwave power. The particle size hassignificant effect on the total saponins yield. Under the optimization conditions, totalsaponins extraction yield was up to13.32%. (3) A novel method of ultrasonic-microwave auxiliary aqueous two-phasesynergistic extraction of total saponins from dioscorea zingiberensis C.H.Wright wasproposed taking the advantages of methods mentioned on the utilization of orthogonalexperiment. The influence factors on the total extraction of saponins wereinvestigated, and the optimization conditions were determined, usingultrasonic-microwave auxiliary aqueous two-phase system of ethanol-(NH4)2SO4synergistic extract method. The order of influence factors were as follows: microwavepower>ultrasonic power> microwave time> ultrasonic time> particle sizes. Underthe optimization conditions, total extraction yield of saponins is up to14.85%. It isone of the highest extraction methods for extraction saponins, the extractionefficiency was improved dramatically.(4) The researches of separate and purify the main ingredients of total saponinswere carried out. And the products obtained were characterized by FTIR andES(+)-MS techniques, etc.
