The Effect Of Three Kinds Of Chinese Herbal Compound On Growth And Immunity Of Carp (Cyprinus Carpioio)

Effects of three kinds of Chinese herbal compound (PESM) on growth, immunity and blood biochemical indicator of carp were studied. A total of 80 fingerlings were divided into four groups. Two repeat were contained in one group. We investigated the growth, immunity and biochemical indices of carp after feeding the carps with PESM continuously for 28 days.The results showed that testing groups exhibited an apparent increase in the weight growth rate compared to control group. It was found that formula AH and AB can increase the concentration of TP, ALB and GLU when bacterial antigen invades. As for the AKP concentration, AH group appears to be increased significantly in all the situations. For the indices of liver function, all these three formulas can reduce the concentration of ALT and AST obviously while only formula AH can also reduce the concentration of LDH. In addition, formula CD can significantly increase the concentration of BUN and CREA as well as the blood sugar concentration; formula AH and AB can give a significant rise to the concentration of Fe in serum.The content of Complement 3 and Complement 4 were both increased significantly by the three kind of PESM compared with control groups. For NO and NOS, the findings showed a larger difference between the organizations. The contents of NO and NOS in serum of Carps feed with Chinese herbal compound were significantly higher than them in control groups . However, in liver, only the contents of iNOS presenced the same change when bacterial antigen were injected. This work demonstrated that these three formula compounds can facilitate the growth of carp as well as improve its immune system.Serum immunoglobulin (Ig) of Cyprinus carpio was also reaserched. It was purified by using saturation ammonium sulfate precipitation followed by Sephadex G– 200 gel column chromatography. Molecular weight and partial characterization of Ig were measured by polyacrylamide gel electrophoresis (PAGE), SDS polyacrylamide gel electrophoresis (SDS - PAGE) and Western blotting. We compared the difference of the contents of serum Ig of Cyprinus carpios which were feed with Chinese herbal compound or no. The results showed that Ig molecular weight of Cyprinus carpios is 807 kD, that heavy chain molecular weight of Ig is 74.8 kD and light chain is 26.1 kD. In western blotting, both heavy chain and light chain of Cyprinus carpio can be detected by rabbit anti. The result of ELISA showed that formula CD significantly increased the concent of serum Ig of Cyprinus carpio.
