| In order to elucidate the metabolic mechanism and distribution of reactive oxygen species (ROS) in the physiological male sterility(PMS).The PMS lines induced by SQ-1,a new chemical hybrid agents (CHA) for wheat .Base on studying the anther morphological to explore the changes in the metabolism and distribution of ROS in the anthers at different developmental stages.The main results are as follows:1.Compare with the control group, most of the pollen cells stay on the latter stage of mononucleate pollen and binucleate pollen stage, and nucleus are more larger; a small number of pollen cells can develop to the trinucleate pollen stage, but they have abnormal sperm nucleus, and some pollen only leave an empty shell.From the latter stage of mononucleate pollen to trinucleate pollen stage, most pollen are crescent-shaped , spindle, and the cytoplasm is rarefied. The results show that the key period of physiological and Sterility Wheat is the latter stage of mononucleate pollen and binucleate pollen stage.2. The treatment group anther tapetum begin disintegrate in the medium stage of mononucleate pollen,and it still exist in the binucleate pollen stage, while the control group begin disintegrate in the early stage of mononucleate pollen. Anther endothecium cells are more obvious thicker of the stripe-like than the control group in the binucleate pollen stage and trinucleate pollen stage ,and cells'shape more clearer. Middle cells arrang in irregular in the Early stage of mononucleate pollen, it still remain exist in the binucleate pollen stage. Description anther wall, including the abnormal development of endothecium, middle cells and tapetum cells may be the reasons of the anther abortion.3. The activities of superoxide dismutase(SOD) in the treatment is lower than corresponding controls except the early stage of mononucleate pollen,but the activities of peroxidase(POD) and catalase(CAT) are all lower than corresponding controls in the all stages. The activities of protective enzymes in both treatments and controls are most different in the late mononuclear and binucleate pollen stage. The same period of O2-. and MDA content also increased the fastest, H2O2 quantitative orientation observed in the same periods, the pollen cells treated with the strongest fluorescence intensity.Analysis, the high level of ROS and low level of protective enzymes are the major abortion factor in pollens.4. Using H2DCFDA to observe distribution of H2O2 in the cellular level, Found distribution of H2O2 in treatment pollen cells have two different types,Most of the cell s'membrane and cytoplasm of pollen present fluorescence,and a litter number of pollen cells fluorescence not only exist in the cell membrane and cytoplasm,and also occur in the inside and outside cell wall. The former one is appeared from the medium stage of mononucleate pollen,and the strongest fluorescence is appeared in the latter stage of mononucleate pollen and binucleate pollen stage, it is almost disappeared in the trinucleate pollen stage .The latter one also appeared in the latter stage of mononucleate pollen and binucleate pollen stage and trinucleate pollen stage . Throught transmission electron microscope and CeCl3 found pollen cells also existe two different types distribution of H2O2 . The first one is CeCl3 precipitation mainly gather in the cell membrane and mitochondria, while the second one is not only gather in the cell membrane and mitochondria, but also appeared in the inside and outside cell wall. And the function of the two different types distribution of H2O2 in the process of pollen abortion need futher study. |
