Lophodermium. spp, causing the falling of pines needles,were investigated thoroughly in pine forests around Liaoning province, including areas in east part of Shenyang and 5 counties and 3 cities of east part of Liaoning province.Using sampling investigate, 50 samples plots were investigated totally. About 100 pathogenic bacteria samples were gained, to carry out arrange, to number, to fractional cultivation, to identify morphology of specific.Some genera of Lophodermium spp which caused needle shed on Pinus tabulaeformis Carr. in east part of Liaoning province were studied. According to the viewpoint of classify by Cannon&Minter (1986), three species of Lophodermium.spp and two var species of Lophodermium.spp were got.They were Lophodermium conigenum(Brunaud)Hilitz, Lophodermium pinastri(Schrad)Chev,Lophodermium nitens Darker,Lophodermium sp.l,Lophodermium sp.2. and Cyclaneusnla minus(Butin)Dicosmo Peredo et Minter. which were cognated with Lophodermium spp. Lophodermium nitens Darker is a newly record species on Pinus tabulaeformis Carr.Used the analysis method of molecular biology——RAPD (randomly amplified polymorphic DNA ), twenty finger printing of strain were got. Analysis the blood relationship in interspecies, fractionate four group, two uncertainty species——Lophodermium sp.1, Lophodermium sp.2 were discussed. Lophodermium sp.1 can polymerize with Lophodermium conigenum, Lophodermium sp.2 can polymerize with Lophodermium pinastri. Variability of different environmental factors were identified. Because of the differences of region, the strains of interspecies to bring about invariably heredity differentiate. The genetic relationship between Cyclaneusnla minus and Lophodermium. spp were discussed. One strain of Lophodermium sp.1 and two strains of Lophodermium sp.2 were sequenced ,each sequence of Lophodermium sp.1 and Lophodermium sp.2 is similar with AY247753.1 and AY515319.1 in GenBank. Showed that the method of molecular biology can combine with morphology appraisement well. Refered to the study methods of forest disease and method of pathogenicity of common phytopathology, to seperrate and cultivate the strain. According to the rule of Koch. Inoculative method to study the pathogenicity were used, use natural spore and bacterial filament to inoculate on the biennial health needle.Found that evry species had pathogenicity, and it had differences in different species, in different inoculative way.different inoculative situs had different Symptom..Through the research of Lophodermium spp on Pinus tabulaeformis Carr.of the east part of Liaoning Province. The main etiological agent which causing the pinus needle disease were dearly and deeply known. Based on the data means of predecessor,estimated the history of classification,morphology,ecological character,geography distribution and bionomics.Approached the authenticity on variety appraisement of method of molecular biology——RAPD,the rationality of rRNA ITS,Provided date means on Lophodermium spp research.and provided theory means on prevention and cure on produce.
