Polygonatum sibiricum Red., which belongs to Polygonatum Liliaceae, is Perennial herb. It has the medicinal value, edible value and ornamental value in one. The rhizomes are tradition Chinese medicine, which have many uses to treat different diseases. As the P. sibiricum medicinal value was deeply understudied, the demands of P. sibiricum are growing more and more. Wild resources have been far from meeting the market needs. Develop the artificial cultivation of herbs medicine is the main way to solve the shortage of P. sibiricum. However, the artificial cultivation of P. sibiricum usually using tubers as reproductive material, that not only waste product, but also have no benefit for large-scale production. And there is almost not make full use of the huge amount of seeds. It is found that the long period dormancy and low germination rate of P. sibiricum seeds are the great obstacles for artificial cultivation. This experiment take P. sibiricum of Buchang pharmaceutical Co. in Lueyang of Shaanxi for test materials. Studying on water absorption rules, bioassays of endogenesis inhibitors of seed coat, endosperm and embryo as well as seed culture etc. to find out the reasons of seed dormancy of P. sibiricum. And studying on the different substrates, different temperatures and exogenous growth materials etc. to find out a right way to break seed dormancy. In addition, study on the changes of soluble protein content, crude fat content, starch content and soluble sugar content, as well as amylase activity etc. during stratifieation for seeds of P. sibiricum. It hopes that can provide theoretical basis for research aspects such as breaking seed dormancy, promoting seed germination, improving technology of artificial cultivation etc..Results showed that:1. Reasons of seed dormancy of P. sibiricum: First of all, the physiological after-ripening might be the main reasons of dormancy. In addition, P. sibiricum seeds have slow water absorption, and there are endogenetic inhibitors exist in different locations of P. sibiricum seeds might be the other reasons of dormancy.2. Right way to break seed dormancy of P. sibiricum: Different temperatures and exogenous growth materials, different chemical reagents, different substrates and physical methods all can effect on P. sibiricum seeds germination. Sand: vermiculite(1:1) which germination rate is 22%, is the best substrate to germinate P. sibiricum seeds. Difference storage methods can effect on seeds activity, and 4℃sand storage is the best way to store P. sibiricum seeds. 25~30℃changing temperature condition and different concentrations of GA3,IAA and 6-BA can promote germination and enhance the germination rates. Weak acid or base is good for P. sibiricum. seeds, while strong acid and strong base are bad for seeds. In addition, physical methods such as removing seed coats and sand scrape are not suitable for P. sibiricum. seeds to break seed dormancy.3. Study on physiologies changes during stratifieation for seeds of P. sibiricum: P. sibiricum seeds which belong to starch-rich seeds, contain abundant starches and sugars. The general trend of soluble protein content, soluble sugar content and amylase activity are increased. And the general trend of crude fat content and starch content are declined during stratifieation process. This shows P. sibiricum seeds have some physiological and biochemical activities, to provide material conditions for seeds germination.
