The research of cutting and tissue culture techniques for Reaumuria soongorica was done in this study,and a practical asexual reproductive technology was preliminarily established,also as an effective expanding progress,it could provide the technical support for Reaumuria soongorica′s exploring.the results were as follows:1 The cutting techniques of Reaumuria soongorica were explored in this study by testing various factors including ages of hormone types,hormone concentrations,cutting periods and matrixs.The best way for hardwood cutting of Reaumuria soongorica was that cuttings came from the middle and top part of 2 to 4-years-old ortets,cuttings soaked in water for 24h,then in NAA 300ppm solution for 20min before inserting in the pure humus soil in the 20th,March,also ventilation,temperature and humidity must contain rational. Under these contations the survival rate could reach a higher value 53.13% and the rooting rate was up to 59.12%.Obvious synergy was found existing in the quantity and length by ABT treatments,which 300ppm ABT processing cuttings can obtain higher average root number 6.3.2 The adventitious bud regeneration system of induction realization technology was preliminarily retained with the material of aseptic seedlings,by the method of shoot induction culture,which the orthogonal and homogeneous experiment were designed.the results indicated that:soked in 75% ethanol for 4s then 0.15% HgCl2 solution for 8min was the best sterilization method. The optimal conditions for initial culture were on the 1/5 MS agar medium with 0.01 mg·L-1 IAA,and the initiation rate was up to 59.77%,also the browning rate could reduce after repeated connecting.The optimal conditions for bud induction and proliferation were on the 1/2MS agar medium with 0.1 mg·L-1 6-BA and 0.01mg·L-1 2,4-D,while 45d would retain the maximum proliferation effect.the optimal rooting medium was on the 1/2 MS agar medium with 0.01 mg·L-1 IAA and 0.01 mg·L-1 ABT,the rooting rate reached 57.54%,and the average rotting number was 4.87,the average root length was 10.62 cm.the survival rate could reach up to 88.76% by choosing pure humus soil as matrix.3 By adding different concentrations of growth hormones and cytokinins,the callus induction was done to research,finally the technology of regeneration system through callus induction was realized,while the response surface analysis method was adopted.the results indicated that the optimal conditions for callus induction were on 1/2 MS agar medium with 0.1 mg·L-1 CPPU and 1 mg·L-1 NAA,the induction rate was up to 99.57%.The optimal conditions for callus proliferation were on the 1/2 MS agar medium with 0.1195 mg·L-1 CPPU and 1.1 mg·L-1 NAA,and the callus multiplication rate was 4.7414.the optimal conditions for callus differentiation were on the 1/2 MS agar medium with 0.05 mg·L-1 CPPU and 1 mg·L-1 NAA,and the callus differentiation rate was 62.56%,while the differentiation bud was 7.53.In a word,through the asexual reproductive technology experiment it is found that the softwood cutting techniques were difficult to realize,and the hardwood cutting and tissue culture could get the sexual reproductive system.the tissue culture by adventitious bud induction or callus induction was better than the hardwood cutting method.
