Poplar is very important for choosing and using the elite clones as short rotation cycleand fast growing industrial tree in Anhui province. Poplar launched as a special species forForest Quality Improvement Action Plan in Anhui province. In recent year, there are someproblems in poplar cultivation: First, with the lagged of seed-reproduction and seedsproduction management, the varieties is gradually with ageing. Second, introduce newvarieties mixed with local varieties, leading to different names with one things or differentthings with one name. Third, because of wide and draggle management and forest quality,insect harm is heavy. In Huaibei plain, plant diseases and insects pests are intensifying yearby year as a result of single poplar varieties, which seriously affect forestry production.In this research, the fast regeneration system has established from leave explants ofPopulus deltoids cv Nankang3, including plant growth regulator, and gets the best trainingeffect. And the genetic diversity of poplar species in Anhui17cultivated varieties and7introduce varieties, building molecular retrieval table and constructing dendrogram. Theresults are as follow:(1) Screen out disinfection method for the explants of Populus deltoides Nankang3: using75%of the alcohol for8s and0.1%corrosive sublimate for4min.(2) Optimized the fast propagation system of Populus deltoides Nankang3and appropriatemedium each phase. Differentiation media was: MS+6-BA1.5mg/L+NAA0.5mg/L+KT0.1mg/L, Multiplication media was: MS+6-BA1.2mg/L+NAA0.3mg/L+KT0.1mg/L, Rooting media was:1/2MS+IBA0.5mg/L+NAA0.01mg/L.(3) Choose the method of improved CTAB to pick up Poplar genome, Electrophoresis andOD value shows that DNA quality accords with molecular biology of requirements.According to the poplar ISSR-PCR systems of lab establish, the primers and theoptimal annealing temperature are screened.(4) The genetic polymorphism of Poplar varieties is investigated using ISSR molecularmarker. Through10primers, for Anhui main cultivated varieties species in Populus,there are54clearly bands of which polymorphic bands number is67, the percentage ofpolymorphism is80.6%; for24poplar varieties, there are56clearly bands of whichpolymorphic bands number is72, the percentage of polymorphism is77.8%.(5) Respectively established Molecular retrieval table for17main cultivated varieties and atotal of24cultivated varieties by special spectral bands. (6) According to the amplification clips, calculate Jaccard genetic similarity coefficient byNTSYS-pc2.10e analysis software and construct dendrogram. In17poplar varieties,the closest genetic relationship is P.×euramericana ‘Nanlin95’and P.×euramericana‘Nanlin895’, which heritability coefficient is0.9620and genetic distance is0.0387; thefarthest is P. deltoides ‘Nankang3’and P. del.cv.Zhongwo1, which coefficient is0.3773and genetic distance is0.9747.In this experiment, establishment of fast regeneration system of Populus deltoids cvNankang3and molecular retrieval table for24cultivated varieties of Anhui province byspecial spectral bands. There provide scientific basis for poplar varieties to identify andthoroughbred promotion. Meanwhile, there has great realistic significance for constructionlarge-scale fast-growing fertility timber and a natural environment.
