High-density genetic linkage map has been applied to effectively the gene location,map-based gene cloning,QTL mapping the position of genes clear which comtroled the important agricultural characters,molecular marker assisted breeding,Comparative Genomics.In this research a molecular marker linage map was constructed in BC1 population which derived from the backcross of'(P. persica (L.) Batsch)'and'(P.kansuensis)'.At the same time molecular markers linked to rootknot resistance nematode character in peach were detected with SRAP markers.The main results were as follows:1.190 BC1 populations were inoculated artificially separately on potted to evaluate their resistance to Mi in open field.107 progenies were susceptible to Mi. and 83 progenies were remarkable resistant, the result was evaluated by the 2 test for goodness of fit against a 1:1 ratio for the dominant markers.2. The stable and hige efficient SRAP annlysis system of peach was established: SRAP system is also 10-μL volume containing 1.0μL template DNA,2.0 u L 10×PCR buffer(MgCL2),0.6 u L dNTP,0.3μL of each primers(lOmM),1 U DNA Termus Aquaticus and 5.6μL ddH2O.3. Polymorphism of parents was analysis with 208 SRAP primers.The selected primers including 53 SRAP primers with rich polymorphism and steady bands were teseted in progeny.102 SRAP markers were assigned to 8 linkage groups. Whole length of linkage map was 994.2 cM,the average length of 8 groups was 124.3 cM, and average interval between markers was9.6 cM.4. Root-knot nematodes resistance marker (Mi-redkansu) were located in group 5.The distanc of genetic linkage map to Root-knot nematodes resistance marker (Mi-redkansu) was 2.1 cM.5. The 36 F2 progeny verifies the validity.Polymorphism of F2 progeny was analysized with the SRAP primer M3E15.The accuracy rate was 91.7%.It is suggested that the SRAP Primer M3E15 might be used molecular marker selection in peach breeding.
