| Cellulose is the most widely distributed, the most abundant, and generated the highest amount of organic compounds. Currently, the development and utilization of agricultural, forestry and fiber by-products is less than 2% throughout the whole world. In our country, more than 50% of agricultural and forestry wastes burned in vain, wasting the precious natural resources. As a major agricultural province, our province is rich in cellulose resources. The full use of cellulose resources has great economic value and practical significance.On the comprehensive utilization of cellulose resources, Screening to the efficient degradation of cellulose-decomposing strain is the key to the use of cellulose resources. However, for the reasons of low temperatures and long duration in winter in some areas of northern China like Heilongjiang Province, rate of cellulose degradation is very slow. Nowadays, the utilization of cellulose degradation by bacteria is in higher growth temperatures, and low rate of degradation of capacity. It is not suitable for northern China with low temperature. Therefore, Screening to low-temperature conditions suitable for efficient cellulose-decomposing bacteria is the key to utilization of cellulose resources in our province.The author takes cow dung as the sample in this paper, getting 50 strains with cellulose degradation activity after screening at 5℃. Then re-screen with the 50 strains using Congo red medium and get 4 strains with clear transparent cellulose rings. By repeated screening with 3,5 - dinitrosalicylic acid assay (DNS method), there is a high cellulose degradation bacteria J-019, the enzyme activity reached 523 U / ml. Appraised by the Institute of Microbiology, strain J-019 is Escherichia coli.The author makes a preliminary study on the J-019 about its enzyme activity. The author tests the factors influencing the enzyme production, including the incubation time of strains, the initial pH value of medium, and the different nitrogen sources and different inoculum. The results for strain J-019 are as follows. 48h is the optimum temperature for enzyme production; 7 is the optimal initial pH value; peptone is the optimal medium nitrogen; and 4% of the inoculum is optimal.The incubation time, initial medium pH, nitrogen source peptone and inoculation acting as four impact factors were studied by cross tests method of four factors and three levels. The test proves that 48h is the best incubation time, 7.5 is the best pH value, and 0.20% peptone and 4% inoculum are the best results.The comprehensive use of cellulose has a bright future in the agricultural provinces rich in cellulose resources. The results getting from this paper is suitable for the climate condition in our province. It laid a solid foundation for the utilization of cellulose resources. |
