Temporary Immersion Bioreactors (TIBs) is a new device for metabolites analysis and plant micropropagation. Liquid cultures were prerequisite to automation and cost reduction in TIBs.Compared with semi-solid culcuture better tissue seedlings and higher multiplication were got in TIBs. In order to promote the banana cultivation, the experiment banana seedlings (Musa AAA Cavendish var. Williams B6) were used as the material which offered by Guangxi plant tissue culture company. Comparison between the plantlets derived from TIBs and conventional semi-culture method for banana russue culture and the effect of different factors on banana micro-propagation in TIBs were investigated. results are as follows:1.Used the sixth subculture banana seedling as materials, the results showed that high multiplication were obtained in TIBs, the multiplication of TIBs was about 14 which was above semi-culture 2-3 times; The seventh subculture and 100 mL of medium per inoculums resulted in better growth and higher multiplication;30 g/L sucrose concentration was suitable for multiplication and rooting in TIBs;2.0 mg/L 6-BA was better for proliferation and growth; 1.5 mg/L GA and 1.0 mg/L NAA was suitable for banana rooting and growth in TIBs;4min/6h temporary frequency was suitable for rooting, while 4min/3h suitable for banana proliferation.2.Used the sixth subculture banana seedling as materials, used Ms+6-BA2.0 mg/L+NAA 0.1 mg/L+agar 6.0 g/L+sucrose 30 g/L at proliferation stage and Ms+GA1.Omg/L+NAA 1.0 mg/L+agar 6.0 g/L+sucrose 30 g/L at rooting stage as semi-solid medium, TIBs and semi-solid culture formula were the same except that TIBs didn't add agar,50 days later, the results showed that compared with semi-solid culture taller shoot length, higher multiplication, more number of leaves, more number of roots, bigger shoot diameter were obtained in TIBs;the survival rate of TIBs was still taller than tradition semi-solid.3.Research the Photosynthetic performance and physiological activity of banana seedlings in different tissue culture after transplanting 20 days.40days,60 days. The results showed that the Net photosynthesis rate, the stomatal conductance and the Transpiration rate of the two culture methods were gradually increasd as the time goes on. while the intercellular CO2 concentration was declined. Overall,the Photosynthetic performance of TIBs was much better than semi-solid culture:The SOD total activity,POD activity and the chlorophyll content of the two methods culture were also increased as the transplanting time goes on, while the MDA content was declined, the MDA content of semi-solid culture banana seedlings was higher than TIBs, while the SOD total activity,POD activity and the chlorophyll content of TIBs banana seedlings were higher than semi-solid.
