In order to study on the influences of natural salt gradient at the bottom ofEbinur Lake on the variety of bacteria in soil, ten different soil samples from A to Jare taken at the northwest part of Ebinur Lake, a test has been conducted on the totalsalt, conductivity and pH value, the result shows that the pH values of soil samplefrom the inside to the outside are between8.0and9.0, weakly alkaline; the result ofconductivity and soluble total salt in soil has been declining from the bottom to theoutside. Then we select the A,D,G and I that interval distance of four sample points islarge and total salt concentration of soils have large differences and build theconstruction of16S rDNA library and RFLP analysis. The A,D,G and I4points forRFLP analysis of bacterial16S rDNA library, RFLP results displayed clearly differentand proved that salt concentration gradient does have an effect on soil microbialdiversity.We do the sequence of16S rDNA library analysis from the bottom of soil sampleof A point in Ebinur lake sediments, the result show that the bacteria which was fromthe bottom of Ebinur lake sediment is divided into four types, respectively includeBacteroidetes,Proteobacteria,Firmicutes and Acidobacteria, In addition, one OTUscan not be sorted to a known group, and which Proteobacteria and Bacteroidetes aretwo important groups. Research results indicates that the bacteria type from EbinurLake deposition are more rare in Xinjiang, and there are a large number of unknownclass group, the bacteria obtained all are is the Halophilic species,and it contains somebacteria types for photosynthesis in the bottom of Ebinur Lake deposition, both existsbacteria types which can metabolism with carbon compounds. So we can infer thatbecause of without plant and animal in the bottom of Ebinur Lake, different types ofbacteria can maintain a small of ecological environment by exercising different ofphysiological function, to reach energy material metabolism of balance.
