Salt stress is one of main stress which can inhibit growth of plant and reduce the crop yield. Long-term since, how to improve the salt resistance capacity of plant and increase the crop yield under salt stress is always been focus by researchers. Developing salt resistance related genes in plant, thus exploring the salt tolerance mechanism of plant and increase the crop resistance to the high salt stress is the focus on current research. Rice is one of the most important crops in the world, its production has been serious influence by some stress (high salt, high and low temperature). So study the salt tolerance of rice, and further improve the ability of salt resistant in rice is great significance. Vesicle transport related protein Rab family plays a key roles in plant salt resistance. In plants, studies have shown that overexpression of AtRab7, and NtRab7will increased the tolerance of transgenic Arabidopsis and Tobocco (salt stress, low temperature). Nabm MY also found that OsRab7s expression controled by the different external stress, but the function and mechanism in its response to abiotic stressesits is not clear in rice.This experiment was constructed the prokaryotic expression vector of OsRab7to study the impact of OsRab7protein in E. coli anti-salt, high and low temperature. And build transgenic vector of OsRab7, get the positive OsRab7transgenic rice plants, study the growth and development phenotypes (height, seed setting rate, etc.) and its resilience physiological and the change of biochemical indices (Na+, proline concentration, etc.) in transgenic plants under the stress (salt, high and low temperature). Studies the growth phenotype of transgenic plant seedlings under high salt condition (the average growth rate of root growth, etc.), further through observed the injury recovery growth phenotype of transgenic plant seedlings under high salt condition, defined the basic function of OsRab7gene in rice tolerance, and initially revealed the role of the protein in mechanism of rice resilience. The research of Vesicle transport-related genes OsRab7in rice resiliences is support the exploration of the molecular mechanisms of resistance in Rice and make a basic theory and practice to the rice resistance breeding. The mainly results are as follows:1. Constructed the OsRab7prokaryotic expression vector successfully, and making OsRab7to normal expression in Escherichia coli. The resilience experiments of OsRab7transformants show that OsRab7s expression can improve the adversity ability of E. coli (high in salt, low temperature).2. Constructed OsRab7transgenic vector Successfully (justice, antisense, interference, GFP), transgenic experment showed that, we get the To progeny transgenic rice plants with a number of222and the positive transgenic rice plants with a number of196, the transgenic efficiency is88.2%. Make the To progeny transgenic rice plants to reproduce Kayo (Chooses20seeds respectively), we get the Ti progeny transgenic rice plants with a number of37. Salt analysis of transgenic rice showed that OsRab7can improve the effectiveness of Salt resistance in transgenic rice.
