Objective:To study the effects of ginkgo biloba extract(EGb) on nuclear factor kappa B (NF-κB) activity and interleukin-6(IL-6) expression in labor mice uterine smooth muscle cells induced by the agonist of neuromedin B Receptor (NMBR).Methods:Primary cells culture of the mice's uterine smooth muscle. Then test cells and NMBR in the was by immunocytochemistry.Detect the activity of NF-κB and expression of IL-6 by siRNA/phosphorylatio ELISa/realtime-PCR and ELISA.Results:1.To screen the effective concentration of EGb761. (1) Effect of EGb761 on the NF-κB activity within smooth muscle cell:High concentration group compared with the control group was statistically significant (P<0.05). Low and median concentration group compared with controls was not statistically significant (P>0.05). (2) The impact of EGb761 on NF-κB activaty within smooth muscle cells induced by NMBR agonists:Besides the median concentration differences between high concentration and the other groups was statistically significant (P<0.05);differences between median concentration group and controls was not statistically significant (P>0.05);the differences between low concentration and NMB group was not statistically significant(P>0.05). 2.NF-κB activity,IL-6 expression level when transfect siRNA or not to mice uterine smooth muscle cells:(1)NF-KB activity, IL-6 expression level when transfect NMBRsiRNA or not:in the absence of NMBRsiRNA the EGb761+NMB group compaired with controls and NMB group is statistically significant (P<0.05), but is not statistically significant with EGb group (P>0.05).It is a statistically significant after in the presence of NMBRsiRNA(P< 0.05).(2) L-6 expression level when transfect NF-κB siRNA or not:the change luler consist with transfect NMBRsiRNA or not.3.The correlation between NF-κB activaty and the IL-6 expression in the presence or not of NMBRsiRNA intervention:It Appears a positive correlation(before contracted R= 0.914, P<0.01,in the presence of NMBRsiRNA R= 0.912,P<0.01,);4.Grading blocking NMBR or NF-κB expression of IL-6 comparisonIt was not statistically significant both in gene level and protein level (p> 0.05)Conclusion:EGb761 can inhibit NF-κB activity and IL-6 expression in the labor mice uterine smooth muscle cells induced by the agonist of NMBR,the mechanism may relat to molecular interaction of NMBR...
