PurposeCancer-related moratalities have been reduced by earlier detection and more effective treatments,but how about the quality of life of cancer survivors,which becomes a new top reserch. One of the most devastating adverse efforts of cancer treatment is damage to the reproductive system,which induces premature menopause and infertility in many of the hundreds of thousands of women. Early menopause frequently occurs in female cancer patients after chemotherapy (CTx),bone marrow (BM) transplantation (BMT) has been linked to an unexplained return of ovarian function and fertility in some survivors,since 1994.This study used a mice modle induced by chemotherapy to explore the therapeutic effects of BMT on injured ovary.Methods48 mice were divided into three groups at random:normal control group(A, n=16), cyclophosphamide and combination of BMT group (B, n=16), cyclophosphamide group (C, n=16).The mice in groups B and C were treated daily with cyclophosphamide 150mg/kg,3 days,i.p.And then allogeneic bone marrow cells were transplanted into the mice in group B. Eight of the animals in each group were killed 21,50 following onset of cyclophosphamide treatment. Vaginal cytology, the morphology of the reproductive, the ovary coefficient and the amount of follicles were compaired to evaluate the fuction of ovaries. For cell tracking, some of these bone marrow cells were marked with Hoechest 33342 and then transplanted into CTx-treated recipients. The ovaries were detected through fluorescence microscope,48 hours later.ResultsOn day 21,50 following onset of cyclophosphamide treatment,the number of mice with regular estrus and the rate of estrus in groupB,C were lower than those in group A. On day 50 the total days of estrus and the rate of estrus in groupB were higher than those in group C (P<0.05).On day 21 the ovary weight, the uterus weight and the ovary coefficient in group B,C ere lower than those in group A(P<0.05); the ovary weight and the ovary coefficient in group B were greater than those in group C(P<0.05). On day 50 the ovary weight and the ovary coefficient in group B, the ovary weight,the uterus weight and the ovary coefficient in group C were lower than those in group A(P<0.05).On day 21,50 the amount of follicles in group B,C were lower than those in group A(P<0.05); the amount of follicles in group B were higher than those in group C(P<0.05).Cell tracking showed that bone marrow cells from dornored mice were founded in ovaries of recipients after BMT.ConclusionThe BMT can repair the fuction of the ovary injured by chemotherapy in mice.But the repairment is too weak to completely restore the ovary to it's former condition. The mechanism is complicated, and the long-term effects are not yet to be seen now. Donor bone marrow-drived cells was observed in the receptor's ovary, which was like a primordial oocytes. Bone Marrow Transplantation could be a new potential strategy for the management of gonadal failure and infertility in femal cancer survivors.
