Study Of The Meridian Of Gastrodia And The Influence Of Composition With Uncaria By Using In Vivo Distribution Method

Gastrodia is the tuber of the Gastrodia elata B plants. It tastes sweet and flat, owned by the liver. It is the most important herbal medicine of stabilizing the liver-yang.Chinese meridian theory, means drug has a special or relatively significant role in some parts of the body, to describe the selectivity of drug action for Chinese medicine.(1)This study researched the meridian theory of Gastrodia and the influence of Uncaria by using the vivo distribution method, with the Gastrodia and Uncaria Combination theory of medicine.This study established the pretreatment and detection methods of gastrodin in heart, spleen and lung.And so did HBA in brain.Pretreatment:Removed tissues, homogenated with a certain percentage of saline, adding a certain percentage of methanol to precipitate protein, filtrated, supernatant dried by nitrogen, adding a certain percentage of re-distilled water as solution, through microporous membranes, save to be detected.Detection Method:Heart and lung tissue as mobile phase, acetonitrile-0.03% phosphoric acid (2:98); spleen mobile phase, acetonitrile-1% phosphoric acid (2:98); brain tissue as the mobile phase, acetonitrile-0.03% phosphoric acid (7:93); all tissues'detection wavelength,221 nm; flow rate,0.8 mL/min; column temperature, conventional 30℃; and the injection volume,10μL.(2)This study investigated the Gastrodia meridian theory and the influence of Uncaria before and after compatibility.In this study, two groups of SHR rats fed, a roup of oral administration of Gastrodia Decoction, another group of oral administration of Gastrodia and Uncaria mixed Decoction.Gastrodia effective components were detected by HPLC in a series of time points. Some conclusions were found as follows.After oral administration, Gastrodia will distribute in rat tissues, in decreasing order of volume as:heart, liver, kidney, spleen, lung and brain(Note:The effective components of the brain as HBA, and other organs as gastrodin). This confirmed Gastrodia will enter the major organs of the liver meridians after oral administration. This also confirmed the correctness of the theory of Gastrodia meridian from the perspective of Biodistribution.Gastrodia distribution volume or trend decreased in the liver, heart, kidney and spleen tissues after compatibility, in which the liver and heart were significantly decreased; The brain tissues had a significantly increasing on distribution of HBA. This showed that Uncaria can influent on Gastrodia Biodistribution and to some extent proved the correctness of the theory of compatibility. Combined with traditional Chinese medicine and modern pharmacological knowledge of pathogenesis, increase of the Gastrodia distribution confirmed that Uncaria can upward Gastrodia into the brain directly and strengthen the therapeutic effect of Gastrodia.
