Objective:To investigate the migration and invasion effect of Cerulenin on human osteosarcoma U2-OS cells in vitro.Methods:The U2-OS cells were treated with different concentrations (0,1,5.0μg/mL) of the fatty acid synthase (FASN) inhibitor cerulenin. RT-PCR was applied to detect the expression of FASN mRNA in osteosarcoma U2-OS cells. Cell scratch test was used to measure the migration of the U2-OS cells, and the cell invasion was detected by transwell chamber assayResult:The expression of FASN mRNA was significantly reduced after treated with cerulenin.Transwell assay showed that the U2-OS cells treated with cerulenin 0,1,5μg/mL for 24h,the number of the cells invasing to the membrane were 107.73±2.98,87.33±3.13,52.4±2.74 (P<0.05).Cell scratch test showed that the cells migration were significantly declined by cerulean.Conclusion:Cerulenin down-regulating the mRNA of FASN can inhibit the migration and invasion of human osteosarcoma U2-OS cells in vitro.
