Objective To investigate cell culture of the human pituitary adenomas in virto and its correlated invasive pathogenesis of pituitary adenomas.Methods50 Pituitary adenomas specimens removed in our department were collected, all specimens were divided into invasive group (20eases) and noninvasive group (30eases) by adopted Hardy- Wilson and Knops standard.Conventional DNA ploidy analysis and chromosome G-band cytogenetics karyotype analysis were applied for chromosomal alteration. The electron microscope, immunohistochemistry and photochemical method were also used to detect ultrastructure, protein expression and hormone secretion of supernatants.Result1,The chromosome deletion involved 1,3 and 18. There were one chromosome in 4,6, two chromosomes in 2,8,9,11,12,13,15,19,20,22, three chromosome in 5,16, four chromosome in 14 and a fragile site on the X chromosome.2,Electron micrograph revealed exaggerated filopodia enlarged nucleous rough endoplasmic reticulum, golgic secration particles of sparse,margination of nucleolus and hypertrophy of mitochondrioa with sparely excreted granules.the expression of prolaction were evaluated by immunohistochemistery using ployclonal antibody in its derived cell-F cultured in vitro.3,The prolactin hormone of cellular supernatant of passages 5,15,20,25,30 in experiment group were (2.97±0.08)ng/ml, (2.91±0.97)ng/ml, (0.92±0.30)ng/ml, (1.12±0.50)ng/ml, (0.62±0.17)ng/ml respectively and had significant differences with control group(all P<0.05).4,The immunohistochemistery assay showed that human prolactin protein staining was positive while other hormones staining were negative in human non-functioning pituitary adenomas derived cell-F.5,Pituitary adenomas culture in vrtro electron micrograph revealed exaggerated filopodia enlage nucleolus, rough endoplasmic reticulum golgi secretion particle of sparse margination of nucleolus and hypertrophy of mitochondria with sparely excreted granules.6,It showed an epithelial -like morphology with large cell nuclei and abundant cytoplasm.Electron microscopy of culture cell revealed that free cell surfaces were raised multiple filopodia, Nuclei contained margination and enlargement of nucleolus. The cytoplasm contained mitochondrial hypertrophy and sparely granulated.Conclusions:1. The chromosome of pituitary adenoma with in viro was Karyological characteristics of the chromosome.2. DNA ploidy analysis shows that is Karyological characteristics for direction to develop.lt is relation to pituitary adenoma for secretion and proliferation function of pituitary adenoma.3. Culture in viro of the pituitary adenoma cell lines with neuroendocrine features, which tumor central area of chromosome DNA might fracture and damage repair enzymes would be to suppress, its expression will to reinforce, Chromosomal Fragile Site has increase to suffer form adenoma for the risk carriers to linker tuomrigenesis4. the immunohistochemistery assay showed that human prolactin protein staining was positive while chromosome of pituitary adenoma has change lead to uncontrolled proliferation of tumor cells and to linker histone modification enzymes cyclin family, the close relationship5.putiutary adenoma in viro with monoclonal...
