Goal:This experiment takes the Chinese medicine diagnosis and treatment theory as the instruction, and carries on comprehensive and standardized research on traditional Chinese medicine dandelion and the dandelion extraction using modern advanced science and technology.Material and method:Use the orthogonal test design to determine the optimum extraction craft for traditional Chinese medicine dandelion; use the highly effective liquid phase chromatography to conduct the quality specification research on dandelion medicinal materials and dandelion extraction, establishing the fingerprint icon for dandelion medicinal materials and dandelion extraction.Results:1. The optimum extraction craft conditions for traditional Chinese medicine dandelion determined by the orthogonal test design is:weigh the dandelion medicinal materials, add water and decoct them three times. Firstly add 20 times of water and decoct the materials for 3 hours; secondly add 15 times of water and decoct the materials for 2 hours; thirdly add 10 times of water and decoct the materials for 1 hour. Then filter and mix the filtrate. When the filtrate pressure reduces and concentrate to the relative density of 1.20 (65℃), add the ethyl while stir, to make the alcohol quantity reach 50%. After leaving it for 12 hours under 20℃, take our the liquid supernatant. When the filtrate pressure reduces, recycle the ethyl alcohol and concentrate to certain amount, heat and concentrate (100℃), reduce pressure and dry (60℃,0.03MPa), make into paste and smash, and finally get the traditional Chinese medicine dandelion. 2. Use the highly effective liquid phase chromatography for content measurement of dandelion medicinal materials and dandelion extraction indicative components. Its linear relationship is good, and the accuracy, stability and duplication and sampling returns-ratio meet the standards. 3. Use the highly effective liquid phase chromatography to establish the fingerprint icon for dandelion medicinal materials and dandelion extraction, and preliminary establish the distinction method for dandelion from different origins.Conclusion:Through comprehensive and standardized research on traditional Chinese medicine dandelion, determine the optimum extraction craft for dandelion medicinal materials for medicine's, perfect the quality standards for dandelion medicinal materials and dandelion extraction, and simultaneously establish the fingerprint icon for dandelion medicinal materials and dandelion extraction, thus making the research on traditional Chinese medicine dandelion scientific and accurate as well as perfect and unified, which provides the reference for further research and development on traditional Chinese medicine dandelion.
