Malignant lymphoma (ML) is derived from the human immune system cells and their progenitor cells and malignant disease. According to clinical and pathological features,ML can be divided into two types:Hodgkin's lymphoma (HL) and non-HodgKin lymphoma (NHL). The incidence of ML is increasing year by year in China. ML often occurs in young adults, and it is seriously threatening human's health and life. At present comprehensive treatment is one of principal therapeutic methods, which palys very important role in improving therapeutic efficacy and prolonging survival.As hematopoietic system disease, NK/T cell lymphoma belongs to non-hodgkin's lymphoma, which accounting for 2% to 10% of all malignant lymphoma. Its pathogenesis, diagnosis and classification is very compliment. Owing to onset occult, difficult for early diagnosis, high invasion, rapid progression, short surviveal and poor prognosis features, the disease threats to human health seriously. Chemotherapy is one of the effective treatments for NK/T cell lymphoma, but drug resistance is the main restriction factor for clinical treatment.At present, there is no preferred treatment perscription for NK/T cell lymphoma,so find the preferred chemotherapy drugs is very important. In recent years, the sensitivity of chemotherapy drugs in vitro has been widely used for cancer research, especially in terms of solid tumors.Drug sensitivity test of tumor cells in vitro have some methods that includes colony formation assay,short-term primary culture cytotoxicity test (MTT, CCK-8 etc.), radioactive labeled DNA precursor incorporation and ATP sensitivity fluorescence detection method. In these test method, MTT method is widely used in the study of various malignant tumors.The CCK-8 test method has the following characteristics:rapid detection, high sensitivity, good accuracy, good repeatability. Which can help us establish the standard treatment through drug sensitivity test in vitro.Currently, NK/T cell lymphoma resistant to most chemotherapy drugs in clinical, so the effective of treatment is poor. The multidrug resistance is the major problem in the chemotherapy of NK/T cell lymphoma. High expression of mdr-1 is an important reason for generate multidrug resistance. Nowadays, the most common technology in this field is RT-PCR. RT-PCR provides a rapid and sensitive method for the analysis of gene expression.It can be used for the detection or quantitative of expression information. In addition, this technology can also be used to detect the differences in gene expression.Overall,RT-PCR is more sensitive and easier than other RNA analysis technology.OBJECTIVE:To observe the effects of cisplatin, carboplatin, oxaliplatin, lobaplatin on human NK/T-cell lymphoma cell line SNK-6 and the expression of multidrug resistance gene 1 (mdr-1) of SNK-6 cells.METHODS:The SNK-6 cells treated by platinum and proliferation was detected with CCK-8. The cell morphology of SNK-6 was detected by inverted microscope, and then 50% concentration of inhibition was calculated. The expression of mdr-1 gene of SNK-6 cells were detected with reverse transcriptase polymerase chain reaction(RT-PCR).RESULTS:1 The different doses of the platinum can remarkably inhibit the proliferation of SNK-6 cells with dose dependency at different levels, and the IC50 of Cisplatinum,Carboplatin,Oxaliplatn,Lobaplatin is 5.737±0.465ug/ml,39.673±3.825ug/ml,11.151±1.397ug/ml,1.527±0.326ug/ml respectively.2 The expression of mdr-1 gene in the SNK-6 cell by RT-PCR.CONCLUSIONS:1 The different doses of the platinum could inhibit the proliferation of SNK-6 cells, but while there were expression of mdr-1 gene in the SNK-6 cell, We hypothesized that the platinum drugs may not depend on mdr-1 and can be used to treat NK/T cell lymphoma.2 when we do drug sensitivity test of the suspension cells,we can use cck-8 method.
