Objective:l.To establish the TNBS-induced SD rat ulcerative colitis (UC) model and the bifidobacterium (Bf) prevention and cure model.2.To investigate the effects of bifidobacterium (Bf) on the gene expression of TLR2,TLR4 and NF-кB in the intestinal mucosa of rats with ulcerative colitis (UC).Methods:1.Animal group division and model set up:A total of 60 SD rats were randomly divided into three groups:blank control group, model control group and Bf group. After adaptive feeding for 7 days as convention,UC was induced by 2,4,6-trinitrobenzene sulphonic acid (2,4,6-TNBS) and 100% alcohol (1:1, V/V) in the model group and Bf group following the methods reported by Morris, the control group were not be treated.. The rats of Bf group were given Bifidobacterium by oral gavages after the models were set up. This treatment in the Bf group was lasted for 3 weeks.2. Three weeks later, all the rats were killed. The conditions of all rats were observed and the intestinal epithelial cells'total RNA was extracted.3.The levels of TLR,TLR4 expression were detected by RT-PCR.4. The expression of TLR4,NF-кB was determined by immunohitstochemical staining.Results:①Both symptoms and the lesions of colonic mucosa in the Bf group were slighter than those of the model group.②The expression of TLR2,TLR4 and NF-кB in the model group were significatly higher than that in the Bf group and blank control group (P<0.01)Conclusion:The gene expression of TLR2,TLR4 and NF-кB can be seen in colonic mucosa of rats with UC. Bf can relieve the inflammation in the intestinal mucous membrane by inhibiting the gene expression of TLR2,TLR4 and NF-кB.
