Objective:The expression of TLR2,PIK3 CA,AKT gene and protein in the colon tissue of wet-hot intragenous ulcerative colitis(UC)model rats was observed,and the effects of the soup on TLR2/PI3K/AKT pathway cytokine were discussed.Methods:By taking food induction of 2,4,6-trinititisnist sulfonate(TNBS)combined with ethanol composite method to replicate the wet-heat-contained UC rat model,60 Wistar rats were randomly divided into blank groups,model groups,lyine sulfonamide(SASP)Group(1g/kg),low in drug soup(6g/kg),medium(12g/kg),high(24g/kg)dose group,self-molding after the success of the 2 nd day of gastric irrigation 1 times per day to the end of the experiment,21d in a row.Colon tissue was collected to observe DAI scores,general state and colon pathological slicing in rats,and to detect gene and protein expression levels using the reverse transcription polymerase chain reaction(RT-PCR)method and the Western blot method.Result:1.Rat activity status(DAI score): Through the general morphological observation of rats,there were significant differences in each group compared with the blank group(P<0.01),and compared with the model group,the DAI score of the drug soup treatment group and the SASP group were significantly reduced,with significant differences(P<0.05).Compared to the SASP group,there was no significant difference in DAI score(P>0.05).2.Rats colon tissue(CMDI score): Compared with the blank group,the pathological score of the model group was significantly higher,the difference was statistically significant(P<0.01),indicating the success of UC modeling;Obviously,the difference was statistically significant(P<0.01),compared to the SASP group,there was no significant difference in pathology score in the high dose group of drug soup(P>0.05),and the difference in the remaining treatment group was statistically significant(P<0.01).3.Expression of colon tissue m RNA: Compared with blank groups,the content of PIK3 CA,AKT,TLR2 m RNA gene in the model group increased significantly(P<0.01);Compared with the model group,the content of PIK3 CA,AKT,TLR2 mRNA gene in the remaining drug soup group was significantly reduced and the expression decreased significantly(P<0.01);compared with the SASP group,TLR2 and AKT in the soup and high-dose group The difference of m RNA content was not statistically significant(P>0.01);compared with sASP group,the difference in PIK3 CA content in the high-dose group of drug soup was not statistically significant(P>0.01);The difference between the gene content and the medium dose group was not statistically significant(P>0.01),and the AKT gene content and the medium and low dose were statistically significant in the high dose group of the drug soup(P<0.01).4.Expression of colon tissue protein: Compared with the blank group,the content of PIK3 CA,AKT,TLR2 protein in the model group increased significantly(P<0.01)and the degree of activation was high;Expression significantly weakened,with the high dose group,medium dose group,lynostic sulfonium group most prominent(P<0.05,P<0.01);The difference of PIK3 CA protein content was not statistically significant(P>0.05),compared with SASP group,the difference of AKT protein content in the high-dose group of drug soup was not statistically significant(P>0.05);The difference between protein content and medium and low dose group was not statistically significant(P>0.05),and the TLR2 gene content and medium dose were not statistically significant in the high-dose group of the drug soup(P>0.05).conclusion:TNBS-induced wet-hot inner-state UC model rats have strong sensitivity to the intervention of the drug soup,can reduce the expression of TLR2,PIK3 CA,AKT and other related genes and proteins,repair the pathological state of UC rats,effectively suppress the immune response to achieve therapeutic effect.
