The Relationship Between The Expression Of Glucocorticoid Receptors And Glucocorticoid Sensitivity In Patients With Sudden Sensorineural Hearing Loss

Objective:To detect the relative expression of human glucocorticoid receptor(GR)α,βsubtype and GRα/GRβin peripheral blood mononuclear cells (PBMC) of patients with Sudden Sensorineural Hearing loss (SSNHL), to discuss the relationship between glucocorticoid resistances and the SSNHL patients after invalid glucocorticoid treatments and reveal the pathogenesis.Method:According to pure tone audiometry(PTA),all patients were divided into flat type, medium high frequency and low medium frequency in 30 cases of SSNHL (experimental group), and every type was devided into treatment sensitive group, treatment invald group based on the response to the treatment with corticosteroids. Reverse transcriptase polymerase chain reaction (RT-PCR) was used to detect the expression of GRcα,GRβand GRα/GRβin PBMC among 30 patients with SSNHL and 10 cases of alone in patients with deviation of nasal septum (control group)Results:(1)The three types had the expression of GRαand GRβin PBMC, the expression of GRa was found no statistical significance between treatment sensitive group and treatment invald group,and the expression of GRa in the SSNHL was found no statistical significance compared to the control group (all P>0.05).(2) In three types, the expression of GRP in the treatment invald group of SSNHL was signifieantly higher than that in both the treatment sensitive group and control group (all P<0.05).(3) The ratio of the GRα/GRPpositive in treatment invald group is decreased than treatment sensitive group and control group (all P<0.05)Conclusions:In SSNHL patients, the expression of GR subtypes in PBMC is closely related to the strength of GC therapy. The high expression of GRβwill result in the much lower rate of GRα/GRβmRNA that related with poor effect.
