Objective:To genotype 3 single nucleotide polymorphisms (SNP) site g.-460C>T,g.1805G>T,g.3375G>C in the FOXJ1 gene and evaluate the association of these SNPs with asthma susceptibility in Chinese Korean population. Methods:Through the case-control study, collected cases and controls, extracted DNA, SNPs g.-460C>T,g.1805G>T,g.3375G>C was genotyped in 32 asthma patients and 50 healthy controls by using polymerase chain reaction (PCR) and direct sequencing using polymerase chain reaction (PCR) and direct sequencing method. The Chi-square test was used to evaluate the Hardy-Weinberg equilibrium. Allele frequencies and significance values were analyzed by using SPSS 17.0 software. Results:These FOXJ1 SNPs were polymorphic in Chinese Korean population, and the gene frequencies and genotype frequencies between cases and controls were consistent with Hardy-Weinberg equilibrium. The frequencies of genotypes GG GA and AA at g.-460C>T complementary strand were0.75,0.25,0.00 in the asthma patients, while 0.62,0.30,0.08 in the healthy controls; and the allele frequency were0.875,0.125 in the asthma patients, while 0.77,0.23 in the healthy controls. The frequencies of genotypes CC, CA and AA at g.1805G>T complementary strand were 0.75,0.25,0.00 in the asthma patients, while 0.62,0.34,0.04 in the healthy controls, and the allele frequency were 0.875,0.125 in the asthma patients, while 0.79,0.21 in the healthy controls. The frequencies of genotypes CC, CG and GG at g.3375G>C complementary strand were 0.594,0.375,0.031 in the asthma patients,while 0.68,0.28,0.04 in the healthy controls, and the allele frequency were 0.871,0.219 in the asthma patients, while 0.82,0.18 the healthy controls. No statistically significant difference was observed in the distribution of genotype and allele frequency between asthma patients and healthy controls(p=0.217, p=0.094; p=0.426,p=0.16; p=0.763,p=0.541).Conclusion:The results of genotype and allele analysis showed that SNPs g.-460C>T,g.1805G>T,g.3375G>C in the FOXJ1 gene aren't associated with asthma in Chinese Korean population.
