Objective:To investigate the expression of Eotaxin and CCR3 in human nasal polyps and their significance utilizing the immunohistochemical staining method.Methods:39 patients were selected, including 28 cases of nasal polyps group and 11 cases with normal inferior turbinates as the control group. HE staining and immunohistochemical staining (SABA method) were utilized and observation the expression of Eotaxin and CCR3 between the nasal polyps group and the control group, and the statistic analysis was carried out.Results:HE staining showed that the epithelial tissue of nasal polyps were thicken and squamous metaplasia occured. Edema could be found in subepithelial stroma, and the intersitial connective tissue was loose, in which a small amount of ectatic capillaries and glands existed. The cystic dilatation of glands could also be found. A variety of inflammatory cell infiltration existed in the tissue, in which EOS infiltration were primary, showing in the mucosa, submucosa and blood vessels and glands of the surrounding tissues. EOS was mainly distributed in the Lamina propria of nasal mucosa (EOS counting:55.64±15.11). Pseudostratified ciliated columnar epithelium was obserbvted on the mucosa of the control group under which a large number of capillaries and small glands was found, and a few eosinophils could be found (EOS counting:5.27±3.72).The number of EOS in nasal polyps group (EOS count:55.64±15.11) was significantly higher than the control group (EOS count:5.27±3.72), and the difference was statistically significant (P<0.01).The positive expression of eotaxin was found in the cytoplasm, mainly showing in the epithelial cells, blood vessels, endothelial cells and inflammatory cells infiltrated in the submucosal lamina propria. Positive staining of Eotaxin in nasal polyps group showed a heavy brown color and wide area. The heaviest straining color occured in the inflammatory cells infiltrated in the submucosal lamina propria(Eotaxin-positive cells: 69.28±19.48). The expression of eotaxin staining in the control group was weak, and maily located in the mucosal epithelium or the infiltrated inflammatory cells (Eotaxin-positive cells:3.82±2.40).The number of cells with eotaxin positive expressing in nasal polyps group (Eotaxin-positive cells:69.28±19.48) was significantly higher than it in the control group (Eotaxin-positive cells:3.82±2.40), and the difference was statistically significant (P<0.01)Positive expression of CCR3 was found in the cells's membranes. The CCR3 was mainly expressed in epithelial cells and EOS cell membrane infiltrated in submucosal lamina propria (CCR3 positive numbers:55.79±21.60) in the nasal polyps, but the expression of the CCR3 was weak in the control group (CCR3 positive numbers:7.45±5.10)CCR3 expressing in nasal polyps group was significantly higher than the control group, and the difference was statistically significant (P<0.01)The expression of eotaxin and the number of infiltrating EOS was significantly positively correlated (r:0.883; P value:0.00), with a statistical significance (P<0.01). The expression of CCR3 and EOS counts was significantly positively correlated (r:0.800, P:0.00), with a statistical significance (P<0.01) and The positive expression of eotaxin and CCR3 was also a significant positive correlation (r:0.776, P:0.00), with a statistical significance (P<0.01).Conclusions:The expression of eotaxin and CCR3 were obviously inhanced with increasing amonts of EOS in the nasal polyps. The eotaxin and CCR3 maybe involved in the migrationg and accumulation of EOS in nasal polyps and playing impotant function to forming nasal polyps...
