Objective:To study the expression of cyclinDl and survivin genes in Hazak's esophageal cancer and the roles of cyclinDl and survivin genes in the development of esophageal cancer.Methods:The expression of cyclinDl and survivin genes were detected in 30 cases of esophageal cancer tissues and 30 cases of no-esophageal cancer tissues by immunohistochemistry.The expression of cyclinDl and survivin genes were detected in 30 cases of esophageal cancer tissues and 30 cases of no-esophageal cancer tissues by reverse transcription polymerase chain reaction.Results:The results of immunohistochemistry:The positive rates of cyclinD1 protein expression in tissues of esophageal squamous cell carcinoma, dysplasi and normal esophageal epithelium were 40.00%,20.00% and 5.00%;survivin protein expression in tissues of esophageal squamous cell carcinoma, dysplasi and normal esophageal epithelium were 40.00%,10.00% and 0%.the normal esophageal epithelium was lower than esophageal squamous cell carcinoma (P<0.0125). The expression of cyclinD1 protein in tissues of well,moderately and poor differentiated ESCC were 40.00%, 35.71% and 50.00%; P>0.05.The expression of survivin protein in tissues of well,moderately and poor differentiated ESCC were 30.00%,35.71% and 66.66%; P>0.05.The results of RT-PCR:The positive rates of cyclinDl gene expression in tissues of esophageal squamous cell carcinoma, dysplasi and normal esophageal epithelium were 63.33%,70.00% and 15.00%, those in esophageal squamous cell carcinoma, dysplasi were different from those in normal esophageal epithelium (P<0.0125). The positive rates of survivin gene expression were 93.33%,60.00%and15.00%, each of tree groups had statistical significance (P<0.0125). The expression of cyclinDl mRNA in tissues of well,moderately and poor differentiated ESCC were 70%, 64.28% and 64.28%; P>0.05. The expression of survivin mRNA were 90%,92.86% and 100%; P>0.05.The expression of cyclinDl and survivin were not related to the degree of tumor's differentiation.Conclusion The expression of protein was accordant with mRNA on survivin and cyclinD 1, which were higher in esophageal squamous cell carcinoma tissues than in normal tissue. The changes in these gene and protein expressions might be the trigger of the carcinogenesis of cholangioncarcinoma. The expression of cyclinD1 and survivin were not related to the degree of tumor's differentiation.
