Objective To investigate the expression of Transforming Growth Factorβ1 (TGF-β1) in the gastric carcinoma,atypical hyperplasia (low-moderate) of gastric mucosa and normal gastric mucosa tissue and the role of TGF-β1 in the gastric carcinoma development,invasion and metastasis.Methods The expression of TGF-β1 was measured by S-P method of immunohistochemical technique in 56 patients with gastric carcinoma,30 patients with atypical hyperplasia (low-moderate) of gastric mucosa and 20 normal gastric mucosa tissues.We investigated the changes of TGF-β1 expression in three groups and the ralationship between TGF-β1 and clinicopathological characteristics of gastric carcinoma;SPSS16.0 for windows software was used to analyse data.Difference of the data between groups was calculated byχ~2 test or nonparametric test. Significant level of test taking P=0.05,the significant difference was considered when P<0.05.Results1 The positive expression of TGF-β1 was brown granule, TGF-β1 was mainly expressed in cytoplasm,occasionally scattered expressed in nucleus.The positive rate of TGF-β1 expression increased significantly in gastric carcinoma tissues(83.9%) compared with that in atypical hyperplasia (low-moderate) of gastric mucosa tissues(43.3%) and that in normal gastric mucosa tissues,with a significant difference(H =45.46,P<0.01).2 The expression of TGF-β1 in patients with poorly differentiated cancers was significantly higher than those in well differentiated cancers(u=-2.26,P=0.024);The expression of TGF-β1 in serosa invasion group was significantly higher than those in no serosa invasion group(u=-3.47,P=0.001); The expression of TGF-β1 in lymph nodes metastatic group was significantly higher than those in no lymph nodes metastatic group (u=-3.87,P < 0.001); The expression of TGF-β1 in clinical stageⅢ+Ⅳgroup was significantly higher than those in clinical stageⅠ+Ⅱgroup (u=-3.19,P=0.001).The expression of TGF-β1 had no correlation with sex,age and different part of gastric carcinomas(P>0.05).Conclusions1 TGF-β1 probably play an important role in the origin and delepoment of gastric cancer.2 TGF-β1 as a possible precancerous lesion to gastric cancer warning factor.3 The expression of TGF-β1 is closely correlated with the malignant biologic behavior of gastric carcinoma, TGF-β1 may become an effective clinic prognostic index of gastric carcinoma. |
