Objective To investigate endothelial cell derived microparticles from peripheral blood and endothelial injury related cell factors (Ang-1, Ang-2, VEGF and TNF-α) in allogeneic hematopoietic stem cell transplantation to probe the pathogenesis of aGVHD. To prove EMPS released from HUVEC after stimulated by TNF-αin vitro.Methods In this study, 21 patients underwent allo-HSCT,①to detect EMPS which express CD62E-PE from peripheral blood by flow cytometry(FCM).②Cell factors Ang-1, Ang-2, VEGF and TNF-α,which related to endothelial injury were detected by ELISA .③EMPS from HUVEC stimulated by TNF-a (different concentration and several action times)were detected by FCM.Results①FCM method showed that the level of EMPS in aGVHD group at +21d after allo-HSCT is higher than no aGVHD group(P<0.05),the level of EMPS at aGVHD point is lower than aGVHD group at +21d(P<0.05),but there is not differernce with other point.②ELISA revealed that Ang-1 increase obviously at +7d in aGVHD group which compared to no-aGVHD group. aGVHD point group had sharply declined than other groups. Ang-2 of aGVHD group was higher than no aGVHD group at +21d, the disparation had significance(P<0.05). The level of TNF-αat +21d were significantly higher than that of aGVHD point group. The level of VEGF is Lowest at +7d than other points.③EMPS stimulated from HUVEC for 24h are more than other time point. In certain range of concentration of TNF-α(10,50,100, 150ng/ml), FCM showed that there are more EMPS than other concentration at 100ng/ml.Conclusion①The level of EMPS and Ang-2 after HSCT were higher than that before HSCT .They not only reflect endothelial injury of aGVHD but also relate with the occurrence of aGVHD.②Ang-1,TNF-αand VEGF could reflect endothelial injury after transplantation state.
