| Objective: To study the components in the aerial part of Clematis aethusaefolia Turcz. and search for new bioactive components. Separate and purify integrates and to get compounds in pure form. Using spectroscopic methods to establish the structures of the isolated pure compounds.Methods: Air-dried aerial part of Clematis aethusaefolia Turcz. (10.6 Kg) was chipped and refluxed in 75%Ethanol three times. The ethanolic extract was decanted and combined; the solvent was evaporated at reduced pressure and obtain extract 1.53Kg.It will be used to extract the same amount of water, petroleum ether, chloroform, ethylacetate, n-butanol in turn extraction of three separate the solvent was evaporated at reduced press. Every parts were measured by the method of brine shrimp lethality to track bioactivity, and the select the most active part to do next research. Using Silica gel column chromatography to fractionate the crude extracts. And reversed phase preparative HPLC, Sephadex LH-20 to separate and purify integrates and to get compounds in pure form. Using spectroscopic methods including1H-NMR,13C-NMR, HMBC, MS to establish the structures of the isolated pure compounds.Result: After the investigation on extraction of the Clematis aethusaefolia Turcz.,11 compounds were isolated from the Clematis aethusaefolia Turcz. These compounds were characterized on the basis of spectral analysis and the structures were estabilished. There was Caffeic acid (1) ferulic acid (2) Apigenin (3) Luteolin-3'-O-glucoside (4) Quercetin-3-O-glucoside (5) Rutin (6) Kaempferol-3-O-rhamnosyl (1-6) glucoside (7) Apigenin-7-O-(P-coumaric acyl) glucoside (8) Apigenin-6-C-(P-CM) C-glycosides of glucose (9) DIBP (10) dibutyl phthalate (11)Conclusion:The result of our experiment indicated that the solvent and methods of extraction used in this experiment are practicable. Column chromatography, preparative HPLC were employed to isolate and purify the components in the aerial part of Clematis aethusaefolia Turcz. In this experiment, and variety of spectroscopic methods were used to elucidated the structures of the compounds. All the compounds were isolated from this plant for the first time and Apigenin-6-C-6"-(P-CM)-glycoside was a new compound. |
