Chinese medicine is the development of the Chinese nation has experienced a long history of excellent preserved treasure, and is an effective method of preventing and curing diseases. Howener, due to historical limitations, the TCM treatment of Chinese medicine tend to stay in our experience and the description, the lack of scientific testing and validation. The use of modern scientific theory and technical means, especially HPLC-MS is the main instrument used for pharmacokinetics analysis and identity of traditional chinese medicine. Mass and effective information will be captured with MS/MS or MS", and will construct a swift, effective analysis system.Radix et rhizoma rhei, the dried roots of Rheum, which is one of the famous herb widely. The main effect are antibiosis, anti-tumor, anti-hyperlipoidemia, anti-hypertension, antiphlogosis, anti-aging, adjust immunity etc. Ablmosechus manihot var. manihot [Hibiscus manihot Linnaeus] is an important traditional Chinese medicine which belongs to Malvaceae family. The seeds, roots, flowers can be used as ingredients of medicine. Modern pharmacological experiments showed that the flower extract had a cretain therapentic effect on nephritis and myocardial injury. It has been used for the treatment of chronic nephritis, diabetes and kidney diseases in clinical practice. Scutellariae Radix, the dried roots of Scutellaria baicalensis Georgi, which is one of the famous herb widely used in the treatment of pyrexia, jaundice, and hypertension. It acts as a key ingredient in a number of formulae such as Shuanghuanglian oral liquid, Huangqin compound recipes granule, Yinhuang table, etc. they are employed generally.In this paper, there is a fast and efficient analysis and detection methods of the Rheum, Scutellariae baicalensis and Abelmoschus Manihot metabolic studies of multi-constituents and natural product prototype components in vivo; the chemical structure of metabolites by HPLC-MS, MS/MS or MSn and other methods. Including drug sample preparation, animal, dosage and time selection, tissue and body fluid samples, biological sample preparatin, the analysis conditions of HPLC-UV and HPLC-MS", detection of biological samples, figure analysis, structure identification, and. data statistics. Furthermore,18 chemical components in Urine sample after oral administration of the extact of Rheum were detected and elucidated in HPLC-MS total ion current, including 5 prototype constituents and 13 metabolites. The 5 and 3 metabolites were identified in drug-containing kidney tissue and plasma. A systematic phytochemical investigantion of S. baicalensis identified 48 chemical constituents by HPLC-MS/MS.7 new and.41 known compounds were isolated and purified through medium-pressure ODS column, as well as repeated preparative using semi-preparative HPLC. The rat urine after oral administration of the extract of A. Manihot and S. baicalensis were studied systematically, using HPLC-MS/MS. Then,9 chemical components were detected and elucidated in HPLC-MS total ion current of Rat urine after oral administration of the extract of A. Manihot, including 2 prototype constituents and 7 metabolites.2 prototype constituents and 7 metabolites were detected and elucidated in Rat urine after oral administration of the extract of S. baicalensis.Throngh this research, the changes of the medicinal ingredients the Rheum, Scutellariae baicalensis and Abelmoschus Manihot were revealed in vivo, especially the Rhubarb. This study can provide valuable examples and methodology for references on the systematic research of Traditional Chinese Medicine.
