Object: The aboveground resources of Rhubarb have Many records of using In folk. Rheum officinale Baill. as genuine regional drug in Sichuan, the main production in Sichuan ABA, that is disaster area of the five one two Wenchuan earthquake. If we can get very good development and use with the aboveground resources of Rhubarb, not only has the vital significance to protect environment and save resources, but also can promote the ecological development in the disaster areas, and improve the economic benefits of the people in the disaster areas. Through the preliminary study on the chemical composition stem leaf of Rheum officinale Baill, to study the purification process of total anthraquinones in the part on the ground of Radix et Rhizoma Rhei with macroreticular resin, to establish the best technological parameter. And study on the extractive of stem leaf of Rheum officinale Baill with Diarrhea, anti-inflammatory, antioxidant effect, in order to find the evidence that the extractive of stem leaf of Rheum officinale Baill can be used as Rheum officinale Baill as alternative resources. Expand the medicine source, The full development and utilization of biological resources, preserve the ecological environment, lay the foundation for the development and utilization of the aboveground resources of Rhubarb.Method:After accessing to information and detailing experimental in much, we find that the Anthraquinone content in the stem or leaf is relatively close to, and the biomass occupies part of the vast majority of the aboveground resources. Considering the maximum utilization of resources, It was decided to study with both stems and leaves together. Research on the general idea is that, first, The adsorption capacity of four types of macroreticular resins to total anthraquinones in the part on the ground of Radix et Rhizoma Rhei were compared with UV, total anthraquinone were selected as markers, to screening the process condition in the purification process of total anthraquinones in the part on the ground of Radix et Riftzoma Rhei with macroreticular resin. Second, survey the effects of defecation of the normal mice and excess heat model mice after the extractive of stem leaf of Rheum officinale Baill, by acute diarrhea, Carbon method and excess heat model respectively. Survey the effects of Inflammation in mice after the extractive of stem leaf of Rheum officinale Baill, by ear edema method in mice and Granuloma model. Survey the effects of antioxidation activity after the extractive of stem leaf of Rheum officinale Baill, by organic radical and hydroxy radical. Last, column chromatography was used for separation, to obtain compounds that from the ethyl acetate extractby of stem leaf of Rheum officinale Baill.Results:(1) D101type maeroretieular resin has good purification process. The best technological parameter was as follows:the maximum sample quantity was4.54g/g (Chinese herbal medicines/macroreticular resins), pH9, flow rate of1ml/min, liquid concentration was2.16g/ml contained Chinese herbal medicines; evenly adsorbent after add sample, water about5times volume of the macroreticular resins was used to rinse the macroreticular resins, then rinse the macroreticular resins with enough10%ethanol, elution solution of75%ethanol mixed with0.4mol/L NaOH in proportion of1:5, consumption of elution solution was10times volume of the macroreticular resins, the purity of total anthraquinones in the part on the ground of Radix et Rhizoma Rhei reached15.25%.(2) The extractive of stem leaf of Rheum officinale Baill causes of diarrhea in mice to a certain extent, the reduction of the normal mice and excess heat model mice the first defecation time, and increased defecation quantity, and inhibit inflammatory reaction in a certain extent. Morever, it also has a certain inhibition effect on the organic free radical and hydroxyl free radical.(3) Get two monomer compounds isolated from the ethyl acetate extractby of stem leaf of Rheum officinale Baill. After determining physicochemical constants, ’H-NMR and13C-NMR, structure identification of the two are progallin A and quercetin.Conclusion:D101type maeroretieular resin has a good enriching effect on the separation and purification of total anthraquinone in he part on the ground of Radix et Rhizoma Rhei decoction, so this process is feasible. The extractive of stem leaf of Rheum officinale Baill may offer Diarrhea, anti-inflammatory and antioxidant benefits, the anti-inflammatory effect is better than that of underground parts. Through the research of the extractive of stem leaf of Rheum officinale Baill, we try to find evidence of that it may be used as Rheum officinale Baill as alternative resources, lay the foundation for the development and utilization of the aboveground resources of Rhubarb.
