Objective To observe the regulation of Scalp-acupuncture (SA) on the positive cells of BrdU,PSA-NCAM,NeuN in the DG of acute cerebral ischemia/reperfusion injury rats, so as to figure out the impact of SA on the proliferation,migration, and differentiation of NSCs of these rats. Combining the effects of SA on ischemic stroke in clinical, to explore the mechanism of SA working on cerebral ischemia.Method 1.70 Wistar rats were randomized into sham group (10), model group (30) and SA group (30). The rats of model group and SA group were made t.o the model of middle cerebral artery occlusion(MCAO) by suture method, which were divided into 3sets respectively according to ischemic time(7d,14d,28d).So there were 7 sets. SA rats were treated after MCAO experiments, Select top temporal forward slash and slash after the top of the temporal of one side (cross check) of the rat for piercing, choose 25 one-inch "Ruiqi Er" (NATURAL) brand disposable sterile acupuncture needles, quick stab to 15°and needle Depth of about 0.5 inches, followed by Han needle treatment, density wave, frequency 2Hz/100Hz, strength of 2mA, every 20min, ltime each day until death. Each rats were injected bromo deoxyuridine (BrdU) solution into abdomen. NeurologicalⅢ severity score (NSS) was applied for each phase point rats.Last observe the BrdU positive cells and the BrdU/PSA-NCAM positive cells of eachset with immunofluorescence assay(IFA). 2. Collect 80 patients with acute ischemic stroke from acupuncture clinic and ward of Wuhan Hospital of Integrated Chinese and Western Medicine, which were randomly divided into SA group and Xingnao Kaiqiao group. Every group was 40. The former was treated by SA, and the latter was worked on Xingnao Kaiqiao.2 groups were 1 day,6 times for a course of treatment interval of 1 day for treatment of 4 cycles. Then observe the difference between the two treatment methods.Result 1. The NSS of Rats after MCAO has great differences, NSS in sham group is far less than model group and SA group in each phase point (P< 0.01).2. Compared with the sham group, BrdU-positive cells of model group and SA group were significantly increased (P< 0.01), and a clearly difference can be seen about BrdU-positive cells between SA group and model group.3. There was a visible increasing on BrdU/PSA-NCAM positive cells of model group and SA group contrasting with that of sham group (P< 0.01);It had great changed on observing the BrdU/PSA-NCAM positive cells of model group and SA group (P< 0.01)4. Compared with the sham group, BrdU/NeuN positive cells of model group and SA group were significantly increased (P< 0.01 ), and the great change also can be seen between model group and SA group in the time point of 14d and 28d.5. After treatment, On NSS at each phase point:SA group demonstrated obviously declined at 28d comparing with that of model group (P<0.05). 6.Clinically, SA therapy and Xingnao Kaiqiao therapy have a significant effect, both groups showed no significant difference.Conclusion 1. Significant change of neurological deficit can be found in the rats after MCAO, and neurological function has improved greatly after SA treatment.2. After cerebral ischemia in rats BrdU positive cells, PSA-NCAM, NeuN were significantly changed, suggesting that cerebral ischemia can promote the proliferation of NSCs, migration and differentiation.3. Scalp treatment can enhance the regulation of NSCs, suggesting that the effect of scalp acupuncture may be the mechanism of ischemic disease, in order to achieve the purpose of repairing Neurological function.4. Scalp treatment of ischemic stroke clinical disease was effective, it is worth promoting.
