Objective: To definite the prevention and therapeutical effect of scalp acupuncture pretreatment on the cerebral ischemic stroke, and to explore the possible mechanism of scalp acupuncture pretreatment for the treatment of ischemic cerebral apoplexy considering the aspect of the stem cells, the effects of the scalp acupuncture pretreatment on ischemia-reperfusion rats after the proliferation, differentiation of endogenous NSCs, BDNF and b FGF were studied.Methods: 120 selected male Wistar rats for experiment, which were randomly divided into three groups named sham, model and experimental group, respectively. There were 40 rats in each group, which was equally divided into 3, 7, 14, 21 d four time points. Through inserting a piece of nylon thread into the internal carotid artery(MCAO), Before building the internal carotid artery, The group of scalp acupuncture pretreatment, we performed bilateral “bai hui, tai yang” points as stimulation points and stimulated time of 20 minutes consecutively every time, after acupuncture treatment to build the modles, the model group only cause arotid artery, don’t do other deals, The sham besides don’t plug into the wire, the rest operations were the same to the model group; Marked proliferation of cells by intraperitoneal injection of Brdu, respectively in 3, 7, 14, 21 d four time points out to study the subjects. Using nerve symptom scores determine the success criteria fo r the model group, by TTC staining and HE staining for cerebral ischemia reperfusion after brain pathology change of the organization. By the immunohistochemical method to determine the Brdu and Nestin positive cells number, we used the double immunofluorescence method to detect how many NSCs differentiate into n euron-like Brd U/GFAP, Brd U/NSE cells,Though the Immunohistochemical method to observe the changes of BDNF, b FGF, to determine the possible mechanism of NSCs proliferation.Results: 1. Compared with the model group, the neural function defect grade of experimental group reduced significantly(P<0.05) at 7, 14, 21 d observing point. 2. Compared with the modle group, The mount of Brdu positive cells of experimental group increased significantly(P < 0.05). The mount of Brdu positive cells of experimental group began to increase at 3 d, reached peak value at 7 d, and decreased at 14 d. 3. Compared with the modle group, The mount of Nestin positive cells of experimental group increased significantly( P<0.05). The mount of Nestin positive cells of experimental group began to increase at 3 d, reached peak value at 7 d,at 7,14,21 d observing point 4. Compared with the modle group, The mount of Brd U/GFAP, Brd U/NSE positive cells of experimental group increased significantly( P < 0.05). The mount of Brd U/GFAP, Brd U/NSE positive cells of experimental group began to increase at 3 d, reached peak value at 7 d,at 7,14 d observing point. 5. Compared with the modle group,The mount of BDNF positive cells of experimental group increased significantly( P<0.05). reached peak value at 7 d, at 7, 14, 21 d observing point. 6. Compared with the modle group, The mount of BDNF positive cells of experimental group increased significantly( P<0.05). reached peak value at 7 d, Experimental group b FGF expression enhanced obviously, at 3, 7, 14, 21 d observing point.Conclusions: 1. The scalp acupuncture pretreatment significantly reduced the neural function symptom scores after cerebral ischemia and reperfusion rats, and could promote the nerve functional recovery. 2. The scalp acupuncture pretreatment significantly prom oted the expression of Brdu 、nestin and Brd U/GFAP、Brd U/NSE cell after cerebral ischemia reperfusion rats, which proved that scalp acupuncture could promote the endogenous proliferation of neural stem cells. 3. The scalp acupuncture pretreatment promoted the expression of neurotrophi c factor b FGF and BDNF for the cerebral ischemia reperfusion rats, which would be helpful to the recovery of neural function. |
