Objective:To detect the expressive levels of thymidine phosphorylase (TP) and D2-40 in esophageal squamous cell carcinoma and the normal tissues near the tumor;to investigate the characteristics of distribution and density of lymphatic vessel and the correlation with its clinicopathological factors in esophageal squamous cell carcinoma ;to analyze the clinical value of TP and MLD in the diagnosis, treatment and prognosis of esophageal squamous cell carcinoma.Method:40 cases of tumor tissue embedded in paraffin with clinical data and follow-up results were collected from the patients who received operation for esophageal squamous cell carcinoma in Taian Central Hospital between 2009 and 2010.All the cases were not treated with chemotherapy or radiotherapy in preoperative management. Immunohistochemistry was used to detect the expressions of thymidine phosphorylase and D2-40. To analyze the expressions of TP and MLD in clinicopathological characteristics of esophageal squamous cell carcinoma.Results:Immune histochemistry shows that TP mainly expresses in cell syrup, the syrup in positive cells is brown in color.The expressiverate of TP in esophageal squamous cell carcinoma and the normal tissue adjacent cancer had significant difference,respectively(P<0.05). The TP expression was significantly associated with lymph node metastasis,differentiate degree and regional invasion,but not associated with age at surgery,gender,tumor size. D2-40 expresses in the lympgatic vessel is positive and brown in color. The expressiverate of D2-40 in esophageal squamous cell carcinoma and the normal tissue adjacent cancer had significant difference,respectively(P<0.05).The MLD expression was significantly associated with lymph node metastasis,differentiate degree and regional invasion,but not associated with age at surgery,gender,tumor size. MLD was higher in TP positive staining group than that in its negative staining group(P<0.05).Conclusion:MLD was correlated with esophageal squamous cell carcinoma lymphatic metastasis, D2-40 was a better index to detect esophageal squamous cell carcinoma lymphatic metastasis. D2-40 has clinical value to gastric carcinoma diagnosis, relapse and metastasis. Statistical analysis revealed that the expression of TP were higher in tumor group than that in normal control group(P<0.05). TP expression was not correlated with age, gender, differentiation degree(P>0.05); but was correlated with lymphtic metastasis, infiltrating depth(P<0.05).Combined detection of TP and D2-40 expression have showed definitely clinical value on metastasis , estimation prognosis , formulating individualized comprehensive therapy plan in the esophageal squamous cell carcninoma. They might indicate lymphatic nodal metastasis of esophageal squamous cell carcinoma. Computer-assisted image analytical system could be used as a parameter for analysising tumor biological behavior such as cancer differentiation level,depth of invasion and lymph nodal metastasis in esophageal carcinoma.
