ObjectiveDetection of thymidine phosphorylase (thymidine incorporationphosphorylase, TP), and vascular endothelial growth factor receptor3(vascular of endothelial growth factor receptor,3, VEGFR-3) expression inesophageal squamous cell carcinoma (of esophageal squamous cell carcinoma,ESCC) and to explore the correlation of two proteins. Using lymphaticspecific marker D2-40detected microlymphatic and determinated thelymphatic vessel density (microlymphatic density, MLD). The detection of TPand VEGFR-3in esophageal squamous cell carcinoma may provideexperimental basis for molecular target for the treatment of cancer anti-tumortherapy for the TP or joint with VEGFR-3.Methods Tissue samples with detailed clinical data from patients in Taian CentralHospital general thoracic surgery during2010-2011, paraffin-embedded80cases,46cases were male and34womencases. All study patients underwentsurgical resection were not such as chemotherapy, radiotherapy, biological,immunological treatment. Thymidine phosphorylase (TP), vascularendothelial growth factor receptor3(vascular of endothelial growth factorreceptor,3, VEGFR-3) detection, and the lymphatic specific marker D2-40antibody staining in esophageal squamous cell carcinoma and normalesophageal tissue micro lymphatic tube mark, counting the lymphatic vesseldensity (microlymphatic density, MLD), the above staining results were Thecomputer image analysis software image-Pro Plus6.0(IPP6.0)post-processing and analysis, observation of the D2-40, thymidinephosphorylase (TP), vascular endothelial growth factor receptor3(ofendothelial vascular growth factor receptor,3, VEGFR-3) in esophagealsquamous cell carcinoma and normal esophageal tissue expression andclinicopathological characteristics of esophageal squamous cell carcinoma,and then explore the TP, VEGFR-3in the pathogenesis of esophagealsquamous cell carcinoma of relevant role.ResultsTP is expressed in cancer cells cytoplasm after immunohistochemical staining, Observed under the microscope, the cytoplasm of positive cellsshowed brown or tan. In the pathological stage later, high lymph nodemetastasis positive rate and high degree of malignancy of esophagealsquamous cell carcinoma the TP expression in cancer tissues of the positiverate was significantly higher than the earlier normal esophageal tissues andpathological stage, lymph node metastasis-negative rate is low, the differencewas statistically significant (P <0.01).VEGFR-3is mainly expressed in thecancer cell cytoplasm after staining showed brown or tan, VEGFR-3expression and the degree of differentiation of the cancer, lymph nodemetastasis, depth of invasion-related, with patients’ age, gender, tumor sizerelevance. D2-40was mainly expressed in esophageal squamous cellcarcinoma and adjacent tissue micro-lymphatic endothelial cells, stainedbrownish yellow. Lymphatic vessel density (MLD) late in the pathologicalstage, lymph node metastasis positive rate, a high degree of malignancy ofesophageal squamous cell carcinoma was significantly higher than in normaltissues and pathological stage earlier, lymph node metastasis-negative rate islow esophageal scales shaped cell carcinoma, the difference was statisticallysignificant (P <0.01). TP, VEGFR-3expression in esophageal squamous cellcarcinoma of the MLD values were significantly higher than the TP ofVEGFR-3MLD values (P <0.01) lower expression levels of cases; Pearson Correlation analysis in esophageal squamous cells, TP and MLD r=0.698,VEGFR-3and MLD r=0.849, TP, and VEGFR-3, the correlation r=0.686,between any two there is a high positive correlation.ConclusionThe expression TP, VEGFR-3, the MLD in the pathological stage later,more lymph node metastasis and advancer malignant esophageal squamouscell carcinoma are significantly higher than normal esophageal tissues andpathological stage earlier, lymph node metastasislow rates of cancer tissue. Inthe TP, VEGFR-3expression higher esophageal squamous cell carcinoma theMLD values is significantly higher than the lower expression levels of TP,VEGFR-3cases and statistical analysis showed that there is a strongcorrelation between TP、 VEGFR-3and MLD, suggesting that themicrolymphatic vessel plays an important role in infiltration, lymph nodemetastasis and deterioration in esophageal squamous cell carcinoma and TP,VEGFR-3may be involved in esophageal squamous cell carcinomamicrolymphangiogenesis. The detection of TP and VEGFR-3in esophagealsquamous cell carcinoma may provide experimental basis for molecular targetfor the treatment of cancer anti-tumor therapy for the TP or joint withVEGFR-3.
