Relationship Between The Expression Of Survivin And Lymphangiogenesis Of Breast Carcinoma

ObjectiveIn this study, the positive expression rates of survivin and lymphatic vessel density(LVD), Lymphangiogenesis-related factors of VEGFR-3,VEGF-C in breast cancer were detected by the level of protein and molecular biology.We investigate the relationship between survivin and lymphangiogenesis,to make known possible molecular mechanism of lymphatic metastasis in breast cancer,to provide new ideas for the clinical diagnosis and treatment of gastric cancer.Materials and methodsForty-six excision specimens for primary breast carcinoma were collected at Affiliated Hospital of Taishan Medical College from July 2008 to July 2010 in this study. According to the actual situation of clinical surgery we divided breast tumors into turmor-area, nearer-turmor area and farther-turmor area . We detected the expression of Survivin protein,VEGF-C protein and VEGFR-3 protein in three areas of breast cancer tissues by immunohistochemical S-P method,and detected the expression of D2-40 by immunohistochemical method in three areas of breast cancer tissues. and counted microlymphatic density.The expression of Survivin-mRNA,VEGF-CmRNA and VGFR-3mRNA in three areas of breast cancer tissues were detected by the technology of RT-PCR. Then we used SPSS software to analyze their relationships and correlations with clinicopathologic findings.Results(1)From tumor-area to nearer-tumor area and to farther-tumor area,the tendency of Survivin-protein and Survivin-mRNA expression rate were decreased and different. The expression of the tumor area was highest.,and was significantly higher than farther-tumor area(p<0.05). The expressin of Survivin-protein and Survivin-mRNA was significantly correlated with lymph-node metastasis in turmor-area(2)From tumor-area to nearer-tumor area and to farther-tumor area,the tendency of VEGF-C-protein and VEGF-CmRNA expression rate were decreased and different.(3)From tumor-area to nearer-tumor area and to farther-tumor area,the tendency of VEGFR-3 -protein and VEGFR-3mRNA expression rate were decreased .The expression of VEGFR-3 in tumor-area to nearer-tumor area was higher than the expression of farther-tumor area.(4)By statistical analysis, a positive correlation was found between the expression of Survivin and VEGF-C,VEGFR-3 in the three areas of breast cancer(p<0.05).(5)From tumor-area to nearer-tumor area and to farther-tumor area,the tendency of lymphatic vessel density was decreased ,the density of nearer-tumer area was highest. The expressin of Survivin-protein and Survivin-mRNA was significantly correlated with lymphatic vessel density in tumor-area and nearer-tumor areaConclusions(1)The over-expression of Survivin-protein and Survivin-mRNA were found in breast carcinoma, furthermore,if the expression of survivin in turmor-area was positive,there were some levels of surviving expression in nearer-turmor area and farther-turmor area. This suggests that turmor-area is the main site of production of survivin. It plays an important role in the development and invasion of breast cancer.(2)The expressin of Survivin-protein and Survivin-mRNA were significantly correlated with lymph-node metastasis in turmor-area.This suggests that the expression of Survivin may be more prone to lymphatic metastasis of cancer cells; it,s overexpression shows a poor prognosis.(3)By statistical analysis, a positive correlation was found between the expression of survivin and VEGF-C,VEGFR-3 in turmor-area and nearer-turmor area area of breast cancer.This suggests that Survivin and VEGF-C,VEGFR-3 had better synergistic effect with tumor lymphangiogenesis. Survivin inhibits apoptosis of lymphatic endothelial cells and promotes lymphangiogenesis possibly through maintaining and strengthening the promotive function of VEGF-C, VEGFR-3 signaling system in lymphatic endothelial cell .(4)The expressin of Survivin was significantly correlated with lymphatic vessel density in tumor-area and nearer-tumor area, and the increase of lymphatic vessel density of tumor provides lymphatic metastasis with more opportunities.This suggests that survivin may promote breast cancer lymphangiogenesis and lymphatic metastasis by paracrine.(5)Survivin expression has a relationship with cancer lymphangiogenesis, it would provide a new targets in the therapy of anti- lymphatic metastasis.
